人骨髓间质干细胞体外培养诱导成为心肌样细胞的实验研究

摘要 目的 研究人骨髓间质干细胞的体外培养及向心肌样细胞转化的条件。方法 取成人髂骨骨髓 ,分离出骨髓间质干细胞进行培养、传代。观察在培养液中添加诱导剂 5 -氮胞苷 (5 -aza)条件下骨髓间质干细胞的生长和分化。结果 成人骨髓间质干细胞贴壁呈集落生长 ,有成纤维细胞样外观。 5 -aza诱导人骨髓间质干细胞转化为心肌样细胞。结论 人骨髓间质干细胞能够在体外被诱导分化为心肌样细胞 ,是心肌组织工程良好的细胞来源。 Objective To investigate the growth and myocardial differentiation of human bone mesenchymal stem cells (BMSCs) in vitro. Methods The human BMSCs were taken from the ilium of a volunteer were cultured. 5-azacytidine (5-aza) for myocardial induction was added to the culture medium, the proliferation and differentiation of cells were noticed. Results The BMSCs were found grew into colonies with the appearance of fibroblastic cells and differentiate into myocardial cells. Conclusion Myocardial cells can be generated from human BMSCs in vitro,and the human BMSCs may be one of the adequate resources of donor cells for myocardial engineering.
