非酶法消除抗坏血酸对尿酸测定的干扰 被引量:1

摘要 目的 研究非酶类物质 4—羟基 2 ,2 ,6 ,6—四甲基—N—氧—哌啶 (ascorbicacidquencher - 2 ,AAQ - 2 )消除抗坏血酸对尿酸测定的干扰。方法 应用OLYMPUS 10 0 0型全自动生化分析仪检测样品中尿酸浓度。结果①不同浓度抗坏血酸可使尿酸检测结果呈不同程度降低 ;②不同浓度的AAQ - 2消除抗坏血酸干扰的能力不同 ;③AAQ - 2与抗坏血酸氧化酶消除抗坏血酸对尿酸测定干扰能力比较 ,2组所测结果无显著性差异 (P >0 0 5 ) ,相关性r=0 .99;④批内、批间平均变异分别为 2 .15 %和 3.2 9% ,平均回收率 95 .3%。结论 非酶类物质AAQ - 2可消除抗坏血酸对尿酸测定的干扰。 Objective To investigate the value of ascorbic acid quencher-2 (AAQ-2) for nonenzymatic elimination of the interference from ascorbic acid in the uricase-based uric acid determination.Methods OLYMPUS 1000 biochemical analyzer was used to measure the concentration of uric acid in the standard and clinical sample solutions.Results ①The uric acid concentration readings were significantly decreased with supplemented amounts of ascorbic acid in the uric acid standard.②Varied concentrations of AAQ-2 had different quenching effects.(3) There was no significant difference in elimination ability between AAQ-2 and ascorbate oxidase treatments (P>0.05), and their correlation was very good (r=0.995 ).④The average within-run CV and between-run CV were 3.29 % and 2.15 % respectively. The average recovery rate was 95.3 %.Conclusion AAQ - 2 was proven to be a suitable quencher of ascorbic acid in clinical samples.
