目的 :本文对角蛋白 17的序列和辅助性T细胞抗原表位进行了分析预测 ,为一定遗传背景下T细胞介导的自身免疫紊乱引发银屑病这一观点提供了研究基础。方法 :以氨基酸组成、疏水性和亲水性分析以及氨基酸替换和二级结构分析的结果为参考 ,以HLADR限制性预测方法为主 ,进行K17的辅助性T细胞表位综合预测。结果 :与HLADR7相关的T细胞辅助性表位可能在 5～ 15肽段、15 0～ 190肽段、2 70～ 30 0肽段和 315～ 35 5肽段 ;与HLADR4相关的T细胞辅助性表位可能在 1～ 10肽段、5 0～ 70肽段、110～ 170肽段、2 70～ 35 0肽段和 4 0 0～ 4 2 0肽段。结论 :本研究为下一步人工合成多肽或表达相应肽段筛选银屑病反应性辅助性T细胞表位打下了一定的理论基础。

Objective: We analyzed the protein sequence and predicted the helper T cell epitopes in order to lay the foundation for the immunological pathogenesis of psoriasis. Methods: We predicted the epitopes by HLA-DR-restricted approach based on the amino acid composition, hydrophobicity, hydrophilicity, amino acid substitution and secondary structure analysis.Results: The HLA-DR7-restricted T cell epitope regions of keratin 17 maybe exist on the 5-15, 150-190, 270-300 and 315-355 peptide regions; the HLA-DR4-restricted regions: 1-10, 50-70, 110-170, 270-350 and 400-420.Conclusion: [WT5BZ]Our study provided a theoretical basis for the followed study of screening psoriasis-specific helper T cell epitopes by synthetic or expressed peptide regions.
