rhG-CSF改变大鼠局灶性脑缺血预后的初步研究

摘要 目的  研究重组人粒细胞集落刺激因子 (recombinanthumangranulocytecolony stimulatingfactor,rhG CSF)对大鼠局灶性脑缺血预后的影响以及与微管相关蛋白 (macrotubule associatedprotein 2 ,MAP 2 )mRNA表达的关系。方法  用线栓法建立大鼠大脑中动脉缺血再灌注模型 ,观察rhG CSF对死亡率和神经功能评分的影响。用半定量逆转录 聚合酶链式反应 (RT PCR)方法 ,测定rhG CSF用药组和非用药组的MAP 2mRNA表达水平的改变。 结果  比较rhG CSF治疗组与非用药组 :治疗组大鼠 ,局灶性脑缺血再灌注后死亡率显著降低 ,神经功能评分明显改善。治疗组缺血侧脑组织的MAP 2mRNA表达明显提前恢复正常。 结论  一定剂量的rhG CSF可以减轻脑缺血再灌注损伤 ,保护神经元 ,改善功能预后。 Objective To investigate the effects of recombinant human granulocyte colony-stimulating factor (rhG-CSF) on the prognosis and the expression of macrotubule-associated protein 2 (MAP-2) mRNA in rats after focal cerebral ischemia. Methods Focal brain ischemia in rats was performed with the intraluminal filament occlusion. The effects of rhG-CSF were evaluated by mortality rate and neurological severity score (NSS). Reverse transcriptional polymerase chain reaction ( RT-PCR ) method was used to measure MAP-2 mRNA expression in ischemic brain tissue of rhG-CSF group and non-rhG-CSF group. Results Compared to that of non-rhG-CSF group the mortality of rhG-CSF group was reduced significantly and NSS was improved markedly. The expression of MAP-2 mRNA in rhG-CSF group recovered earlier than that of non-rhG-CSF. Conclusion The rhG-CSF may alleviate the brain damage following ischemia-reperfusion and improve the functional prognosis.
