摘要 目的 :研究鸟氨酸脱羧酶 (ODC)基因在结直肠癌中的表达 ,探讨其在大肠癌形成中的作用。方法 :建立测定ODC基因mRNA表达的逆转录多聚酶链反应 (RT PCR)最适条件 ,在半定量水平测定 31例结直肠癌的癌旁正常组织和癌组织中的ODCmRNA表达的相对水平 ,观察其与临床病理学指标的关系。结果 :31例结直肠癌中ODC基因表达较癌旁正常组织高 ,差异显著 (P <0 .0 1) ,ODC基因表达在性别、病理类型之间差异无显著意义 (P >0 .0 5 ) ,在Dukes分期之间差异有显著意义 (P =0 .0 12 )。结论 :ODC在大肠癌组织中表达增高并与恶性程度呈正相关 ,提示ODC表达增加与大肠癌发生、浸润及转移有关 ,为大肠癌病因及诊断和治疗方法研究提供了实验依据。 Objective:To investigate the expression of ODC mRNA in colorectal neoplasms.Methods:Total RNA was extracted from 31 surgically resected specimens of colorectal caricinoma and normal mucosa closed to the lesions.After the optimal reverse transcription polymerase chain reaction (RT-PCR) condition was found,the mRNA expression levels of the ODC gene in the lesions and the normal mucosa were examined semi-quantitatively and the relation between these expression levels and clinical pathological markers was compared.Results:The expression of ODC gene mRNA in lesion of colorectal carcinoma was significantly highter than that in contiguous normal mucosa (P<0.01).Based on the ratio of ODC expression in neoplasm to that in contiguous mucosa,the 31 patients were divided into two groups:over-expression group (12 cases) and high-expression group (19 cases). In the over-expression group the lesions of 7 cases were at the stages of Dukes C and Dukes D;but in the high-expression group,only 2 cases were at the stages of Dukes C and Dukes D,the difference in Dukes' stages between the two groups was statistically significant (P=0.012).Conclusion:ODC gene plays an important role in colorectal carcinogenesis and the up-regulated expression of ODC mRNA is related to the Dukes stages' of colorectal cancer.
