不同细胞系对棉铃虫核型多角体病毒复制的受纳性研究 被引量:1

摘要 用棉铃虫单粒包埋型核型多角体病毒(HaSNPV)分别感染棉铃虫细胞SFE-HA8212(HA-8212),草地贪夜蛾细胞(sf)和中国仓鼠卵巢细胞(CHO)3株细胞系,观察了它们对HaSNPV复制的受纳性。结果表明,HA-8212细胞为受纳性类型:感染细胞出现细胞病变;测定感染细胞上清液中病毒TCID_(50)值表明,病毒粒子不断地形成并释放;以HaSNPV的单克隆抗体MAD_7和多克隆抗体检测出感染细胞中有病毒抗原合成;用^(35)S-Met标记显示有10种病毒特异蛋白(ICSP)分期合成;用标记的病毒DNA作探针进行分子杂交显示细胞中有大量病毒特异的核酸合成。CHO细胞为非受纳型:感染细胞生长正常,无病毒粒子或任何病毒生物大分子生成。Sf细胞为半受纳型:感染细胞破碎,仅在一定阶段有病毒特异核酸合成,但无病毒粒子和ICSP形成。文中讨论了研究细胞系对病毒复制的受纳性的应用意义。 Three different cell lines were infected with Heliothis armigera nuclear polyhedrosis virus ( singly embeded ) ( HaSNPV ) to investigate their per-missiveness for HaSNPV replication. These cell lines are, H. armigera cell line SFE-HA-8212 ( HA-8212 ), Spodoptera frugiperda cell line ( Sf ) , and Chinese hamster ovarian cell line(CHO). The results are as follows. HA-8212 is a permissive cell line that exhitits cytopathic effects typical of NPV infection; formation and release of virulent virus particles by infected cells; synthesis of virai antigens detected by HaSNPV polyclonal an-tibodies and monoclonal antibody MAD7; production of infected cell spe-cific proteins ( ICSPs ); synthesis of virus specific nucleic acid detected by molecular hybridization. CHO cell line is non-permissive and is not af-fected. Sf cell line is semipermissive. Sf celis are disintegrated after HaSNPV infection and synthesis of virus specific nucleic acid is detected at certain stage after HaSNPV infection. Nevertheless, there is no production of virus particles and ICSPs. The practical implications of these results were discussed.
