摘要
目的 :观察愈肝胶囊对HBV转染细胞表达功能的影响。方法 :本研究以乙型肝炎病毒 (HBV )基因转染的人肝癌细胞系 2 2 15细胞作为筛选抗HBV药物的细胞模型 ,观察愈肝胶囊对 2 2 15细胞分泌HBsAg、HBeAg的影响 ;并以四甲基氮唑蓝 (MTT )比色法检测药物对细胞的毒性。结果 :愈肝胶囊对 2 2 15细胞分泌HBsAg和HBeAg有显著抑制作用 ,半数中毒剂量 (TC5 0 )为 1 79mg/ml,对 2 2 15细胞分泌HBsAg和HBeAg抑制率分别为70 81% ,83 96% ;半数有效剂量 (IC5 0 )HBsAg为 0 86,HBeAg为 0 13。治疗指数 (TI)分别为HBsAg 2 0 8,HBeAg 13 76。 结论 :愈肝胶囊在体外有一定的抗HBV作用。
Objective:To investigate the effect of Yugan Capsule on expressing function of HBV transfection cell. Methods: Using the HBV genetic transfection hepatic cell of human-2.2.15 cell as cellular model to screen anti-HBV drugs. To observe the effect of Yugan Capsule on the 2.2.15 cell secreting HBsAg & HBeAg and test it's toxicity on hepatic cell with MTT chromometry. Results: Yugan capsule had markable suppressor effect on the 2.2.15 cell secreting HBsAg, the median toxicity dose(TC50) was 1.79mg/ml, the inhibition rate on HBsAg & HBeAg were 70.81% and 83.96% individually, the median effective dose(IC50) were 0.86 and 0.13, the therapeutic index of HBsAg & HBeAg were 2.08 and 13.76. Conclusion:Yugan capsule had antiviral effect in the extrinsic test.
