流行性出血热病毒感染小鼠脾脏中的细胞毒因子 被引量:2

CYTOTOXIC FACTOR IN THE SPLEEN OF MICE INFECTED WITH EPIDEMIC HEMORRHAGIC FEVER VIRUS

摘要 流行性出血热(Epidemic hemorrhagic fever,EHF)病毒感染小鼠的脾脏匀浆液中存在细胞毒因子,它对正常小鼠的脾细胞、胸腺细胞和腹腔渗出细胞均有细胞毒作用,并对刀豆球蛋白A(ConA)转化的小鼠淋巴母细胞有强烈的杀伤活性。细胞毒活性的高峰发生在病毒感染后第12天。此种脾脏匀浆液在体内外试验中对ConA诱导的淋巴母细胞转化能力亦有明显的抑制作用。感染鼠的其它脏器匀浆液及EHF病毒本身均无此种细胞毒活性。 A cytotoxic factor ( CF ) was detected in the spleen homogenate of mice infected with epidemic hemorrhagic fever ( EHF ) virus ( VISH ). Mice inoculated intracerebrally with 5×105 TCID50 of EHF virus Al6 strain showed the presence of a CF in the spleen homogenate which is cytotoxic to spleen celis, thymocytes and peritoneal exudate celis of normai syngenic mice in vitro. Maximum cytotoxicity is present on day 12 after infection. Furthermore, VISH kills lymphoblasts of mice trans-formed by concanavalin A ( ConA).86.52±15.37% of lymphoblasts were killed when co-cultured with VISH for 24hr, increasing to 98.15±0.79% by 48hr. In addition, VISH suppresses lymphocyte blastogenesis transf-ormation induced by ConA significantly both in vitro and in vivo. No cy-totoxicity was found in homogenates of other organs from infocted mice and spleen homogenate from mice injected introcerebrally with normai brain suspension of suckling mice. EHF virus A16 strain itself also had no cytotoxicity on lymphoblasts. Based on these findings, we sug-gest that the EHF virus-induced CF may play an important role in the suppression of cellular immunity and in the pathogenesis of EHF.
