摘要
以甜樱桃为母本 ,中国樱桃为父本 ,进行种间杂交 ,建立了杂种胚抢救技术体系 ,并对部分杂种进行了鉴定。结果表明 :(1)母本中 ,‘养老’坐果率最高 ,‘养老’在授粉 4周后胚发生败育 ,而‘那翁’与‘红灯’则是在第 3周后败育 ;(2 )最佳萌发与生长培养基为 1/ 2MS +BA 2mg/L +IAA 1mg/L +维生素C 10mg/L ,最佳继代增殖培养基为 1/ 2MS +BA 2mg/L +IAA 1mg/L +NAA 0 1mg/L +维生素C 10mg/L ,最佳生根培养基为 1/ 2MS +IBA 0 2mg/L +维生素C 10mg/L ;(3)S allelespecificPCR。
Interspecific hybridization was carried out with selecting sweet cherry as female parent and Chinese cherry as male parent. Embryo rescue system of hybrids were studied and identification of them was also made. The results showed that: (1)setting percentage was highest when'Elton'was female parent; embryo abortion happened 4 weeks after pollination in'Elton', 3 weeks in'Napoleon'and'Hongdeng';(2)1/2 MS medium supplemented with BA 2 mg/L,IAA 1 mg/L and vitamin C 10 mg/L was the optimal medium for germination and growth, 1/2 MS medium supplemented with BA 2 mg/L,IAA 1 mg/L, NAA 0 1 mg/L and vitamin C 10 mg/L was the optimal medium for multiplication, 1/2 MS medium supplemented with IBA 0 2 mg/L and vitamin C 10 mg/L was the optimal medium for rootage; (3)S allele specific PCR?RAPD and leaf shape analysis showed that they were real hybrids.
