摘要
草莓外植体在不同组合培养基上诱导产生 3类愈伤组织 ,Ⅰ型愈伤组织的再生能力较低 ,Ⅱ和Ⅲ型愈伤组织具有较高的再生能力。Ⅱ和Ⅲ型初生愈伤组织H2 O2 的含量分别是Ⅰ型的 5倍和 9倍 ,O·2的释放速率是其 3倍和 4倍 ,并且具有较高的抗氧化酶活性。在继代培养过程中 ,愈伤组织的形态发生能力下降 ,Ⅰ型愈伤组织逐渐失去再生能力 ,同时所有类型愈伤组织中H2 O2 含量缓慢上升 ,O·2 释放速率较快增长 。
Three types of callus,which present different regeneration capacity,were established in the tissue culture of strawberry.The Ⅱ and Ⅲ types of callus possessed higher regeneration capacity while Ⅰ type of callus had much lower.In the freshly induced calli,the Ⅱ and Ⅲ types of callus had 5 fold and 9 fold higher contents of intracellular H 2O 2,and 3 fold and 4 fold higher level of O [BHDFG1.5mm,WK1.5mmW]· 2,compared with the Ⅰ type of callus, respectively .Also,the calli Ⅱ and Ⅲ had much higher activities of antioxidant enzymes than the callus Ⅰ.During the subculture,H 2O 2 level increased gradually and O [BHDFG1.5mm,WK1.5mmW]· 2 rapidly,while the activities of antioxidant enzymes gradually declined.The ability of expression of totipotency in calli decreased,especially the callus Ⅰ lost the ability after 12 week subculture. The results indicated that the reactive oxygen species might play a dual role in the regeneration of strawberry calli.On the one hand,a certain level of ROS may have a positive effect on the regeneration,and on the other hand, high level of ROS is inhibitory for the expression of totipotency in calli.
