摘要
以油用向日葵新葵杂6号品种为材料,采用不同浓度梯度抗生素处理茎尖、子叶、胚轴转化外植体的方法,研究了不同浓度的卡那霉素和潮霉素对向日葵茎尖、子叶、胚轴不同部位遗传转化外植体的筛选效果。结果表明,卡那霉素对向日葵茎尖、子叶、胚轴不同部位转化外植体的筛选适宜浓度为25~30 mg/mL,潮霉素对向日葵茎尖、子叶、胚轴不同部位转化外植体的筛选适宜浓度为7~10 mg/mL;PCR检测结果显示阳性植株外源基因已导入;不同部位的转化外植体对卡那霉素和潮霉素的敏感性存在一定的差异,但未见明显梯度。
The effects on the different part explants of New Hybrid 6 sunflower with different concentration of kanamycin and hygroycin were investigated. The results indicated that the optimal concentration of kanamycin for selecting the different part transformation explants was 25-30mg/L and adequate concentration of hygromycin was 7-10mg/L. PCR analysis of the kanamycin resistant transformations and hygromycin resistant transformations manifested that the foreign gene was integrated into the sunflower genome. The transformation explants of different part had different sensitivity to kanamycin and hygroycin,but they showed no evident gradient.
