摘要
利用酚抽提法分别提取对照、3%NaHCO3处理 5d和 1 0d的西伯利亚蓼地茎、茎和叶部蛋白 ,用十二烷基硫酸钠—聚丙烯酰胺凝胶电泳 (SDS—PAGE)和分析软件 (Labwork 3.0 )对提取蛋白进行分离和定量分析。结果表明 ,地茎蛋白图谱在 34.1和 2 5 .0× 1 0 3D位置处出现了两条明显受正向调控的蛋白条带 ,茎部蛋白图谱在 5 8.1× 1 0 3D、47.8× 1 0 3D和 9.6× 1 0 3D位置处出现三条正向调控的条带 ,叶组织蛋白图谱在 2 5 .0× 1 0 3D处出现一受正调控的条带。除地茎和叶组织在 2 5 .0× 1 0 3D处同时出现受正向调控的蛋白条带外 ,盐处理后其它组织间表达受正向调控的蛋白条带间存在差异 。
Proteins in under-ground stem, stem and leaf tissues of Polygonum sibiricum which were watered with 3% NaHCO 3 for 5 days, 10 days and with tap water for control were extracted by phenol extraction method and separated and analyzed by SDS-PAGE and the related software labword version 3.0. The results show that two bands were up-regulated in the map of the under-ground tissue with the molecular weights of 34.1×10 3D and 25.0×10 3D, three bands were up-regulated in the map of the stem tissue with the molecular weights of 58.1×10 3D?47.8×10 3D and 9.6×10 3D and one band was up-regulated in the map of the leaf tissue with the molecular weight of 25.0×10 3D. Apart from the same band at 25×10 3D position on the maps of the under-ground stem tissue and the leaf tissue, the bands at other positions on the maps of three parts of the plant which were up-regulated were different. The diversities of the patterns of protein expression in different parts of the plant are illuminated by the results that were gained.
