摘要
目的探讨CD105在结直肠癌中的表达及其作为肿瘤新生血管标记物的临床价值。方法采用鼠抗人CD105单克隆抗体(MAb),应用免疫组织化学技术检测48例结直肠癌手术切除标本及48例对照组织的微血管密度(MVD),并以Ⅷ因子相关抗原(F-8RAg)及血管内皮生长因子(VEGF)的表达作为对照研究。结果(1)MVD及VEGF在结直肠癌组织与对照组织间表达的差异具有非常显著性意义(P<0.001,P<0.01);(2)以CD105和F-8RAg测得的MVD差异具有非常显著性意义(P<0.001);(3)VEGF在结直肠癌的表达与CD105的表达呈显著正相关(P<0.01),而与F-8RAg的表达无关(P>0.05)。结论CD105在结直肠癌组织新生血管有良好表达,可以作为结直肠癌新生血管有价值的标志物。以CD105测得的MVD可以被认为是与肿瘤生长、转移及预后有关的一种重要指标。
Objective To determine the expression and clinical significance of CD105 as a marker of neoplasm related angiogenesis in colorectal cancer. Methods Immunohistochemical staining with anti CD105 monoclonal antibody,anti F 8 RAg and anti VEGF MAb was used to determine CD105 expression and MVD in tissue specimens from 48 cases of colorectal cancer and 48 normal control specimens. Results The MVD marked by anti CD105 and anti F 8 RAg and the positivity of VEGF in colorectal cancer tissues were significantly higher than those in control tissues(P< 0.01 or P< 0.01). The MVD marked by anti CD105 in colorectal cancer tissues was significantly higher than that marked by anti F 8 RAg (P< 0.001). VEGF expression was significantly correlated with CD105 expression,but not with F 8 RAg expression. Conclusions CD105 is expressed highly in the vascular endothelia of microvessles of colorectal cancer, and its expression can be used as an valuable marker of angiogenesis. The MVD marked by anti CD105 is associated with tumor growth, metastasis and prognosis.
