摘要
为探讨非特异性损伤因素对排斥反应所造成的影响 ,建立仓鼠对裸小鼠心脏移植模型 ,分别在手术当天 ,术后 10 0d及 2 70d重建受者免疫系统 ,观察仓鼠移植心的存活及移植心组织内热休克蛋白 70的表达水平。发现随术后免疫重建时间的推移 ,移植心组织内热休克蛋白 70的表达则逐渐降低 ,而受体排斥移植心所需时间逐渐延长。提示包括非特异性损伤(手术、缺血、感染等 )在内的各种因素能在相当长的时间内参与诱发移植排斥反应。
To investigate the influence of the non-specific damages on the rejection reactions,a transplantation model with immunodeficient recipients was established and splenocytes or purified T cells were adoptively transfused at 0,100 and 270 post operative days after xenotransplantation to reconstitute the immune system of the recipient mice.The mean survival time (MST) of xenografts was assayed by daily inspections,and the expression of HSP70 in cadiac graft tissues were detected by Western blotting analysis. It was found that with prolongation of the time in immune reconstitution,the rejection time was gradually prolonged with a graduate reduction of the HSP70 expression in transplanted cardiac tissues. It concludes that the nonspecific damage factors,such as operation,ischemia and infection,participate in the induction of cardiac xenograft rejection for a fairly long time.
