摘要
目的 探讨结直肠癌HPV16型感染与核因子 κB(NF κB)活化的关系。方法 应用凝胶电泳迁移率分析 (EMSA)检测 5 0例结直肠癌、30例结直肠腺瘤和 2 0例正常大肠组织核因子NF κBDNA结合活性 ,并用多聚酶链反应 (PCR)和southernblot检测HPV16型DNA。结果 结直肠癌、腺瘤分别与正常结直肠组织比较 ,NF κBDNA结合活性和HPV16型DNA阳性率均明显增高 ,差异有显著性 (P <0 .0 5 ) ;结直肠癌和结直肠腺瘤比较 ,NF κBDNA结合活性和HPV16型DNA阳性率差异均无显著性 (P>0 .0 5 )。HPV16型DNA阳性和阴性结直肠癌患者比较 ,NF κBDNA结合活性差异有显著性 (P <0 .0 5 )。结论 NF KB活化参与HPV16型在结直肠癌的致癌过程。
Objective To assess activation of nuclear factor-κB (NF-κB)and Human papillomavirus (HPV) type 16 DNA in human colorectal carcinoma.Methods Both detection of activation of NF-κB by electrophoretic mobility shift assay(EMSA) and HPV type16 DNA by polymerase chain reaction (PCR) were performed in tissue of the patients with colorectal carcinoma (50 cases),adenoma(30 cases)and normal colorectal tissue (20 cases ).Results The results showed both NF-κB binding activity and the positive rates of HPV type16 DNA were significantly higher in patients with the colorectal adenocarcinoma(or adenoma)than the normal colorectal mucosal samples( P < 0.05 ),but no clearly higher in patients with the colorectal adenocarcinoma than the colorectal adenoma ( P > 0.05 ). NF-κB binding activity in patients with HPV type 16 DNA positive were significantly higher than those in patients with HPV type 16 DNA negative( P < 0.05 ).Conclusion Activation of NF-κB is related to carcinogenesis of HPV type 16 in patients with colorectal adenocarcinoma.
