Hypoxia-inducible factor 1(HIF-1) is a heterodimeric transcription factor that functions as a master regulator of mammalian oxygen homeostasis. Under hypoxic conditions HIF-1 becomes activated and up-regulates downstream target genes. HIF′s function of oxygen homeostasis is achieved by regulating about 50 target genes which encode proteins that mediated the adaptative responses of the cell to hypoxia. Therefore, HIF-1 plays a pivotal role in the process of hypoxia signal transduction, being of very important significance in physiology and pathophysiology. In this review, some of the latest advances of research in HIF-1 structure, activity regulation and target genes are summarized.
