摘要
目的 :探讨人胶质瘤发生与多瘤病毒SV40、JCV和BKV的PCR扩增情况 ,并对扩增产物做了基因测序和序列分析。方法 :采用PCR方法检测 94例胶质瘤和 18例正常脑组织中SV40、JCV和BKV的基因片段 ,并对基因片段做了克隆测序和序列分析。结果 :SV40和JCV序列与GenBank序列完全同源 ,三条BKV序列有两条与GenBank公布的序列有部分硷基不同 ,但氨基酸序列分析结果则相同。结论
Objective To investigate the relationship between human glioma produced and polyomavirus(SV40,JCV and BKV). Methods PCR was used to detect SV40,JCV and BKV in 94 glioma samples and 18 normal brain tissue samples, and the gene clone sequences were determinated and analysed.Results SV40 and JCV sequence amplified and the sequence in GenBank were 100% homologous. The basepairs were different between 2 of 3 BKV clone sequences and the sequence in GenBank, and the other was 100%.But the 2 different basepairs lied in the codon, which did not influence the translation of amino acid. The amino acid sequences of the 2 DNA different sequences were 100% same as the sequence in GenBank.Conclusion Subtypes of polyomavirus can lead to human glioma.
