摘要
目的 :建立树突状细胞的体外扩增方法并研究树突状细胞的功能。方法 :根据细胞的黏附性不同 ,建立了一整套简便的树突状细胞的体外扩增方法 ,并利用HE染色和FACS研究了其生物学特征。结果 :体外培养 3d后 ,可见增殖性细胞集落 ,体外培养第 7d ,收集悬浮细胞即为DC。倒置显微镜下见 :细胞呈树突状、伪足状突起 ,HE染色核偏位 ,嗜碱性。流式细胞鉴定为髓系DC(CD11c+ ) ,高表达MHCⅡ类分子 (Ia)及共刺激分子 (CD40 ,CD80 ,CD86)。结论 :此实验制备的树突状细胞具有髓系树突状细胞的生物学特性 。
Objective To set up a series of method to generate dendritic cell(DC) and study its function.Methods According to the difference of cells's adhesion,a series of method were set up to generate DC and the biological characteristics were studied by HE staining and FACS.Results The proliferating clusters were found in 3 days and DC was collected in 7 days.The cells showed branching like and pseudopod like under inverted light microscope.The bias nucleus of DC were basophilic after HE staining.These DC belonged to myeloid lineage of DC(CDllc +) and high express MHCⅡ(Ia) and co stimulatory molecules (CD40,CD80,CD86)by FACS.Conclusion The DCs generated have cytobiological characteristics of myeloid lineage of DC,and it will be widely used in the clinical study of and experiment.;
