摘要 目的 了解嗜血杆菌鉴定中X、V因子测定的各种影响因素及可用于嗜血杆菌鉴定的卫星试验的细菌种类。方法 用 4种不同的琼脂平板 ,比较不同培养时间和CO2 环境对嗜血杆菌X、V因子需求试验结果的影响 ,同时比较不同菌种在各种琼脂培养基上对嗜血杆菌琼脂卫星试验结果的影响。结果 X、V因子需求试验结果表明 ,脑心琼脂效果最好 ,其次为营养琼脂 ,M H琼脂结果与脑心琼脂相比差异有显著性 (P <0 .0 1) ;CO2对试验结果有影响 ;培养 4 8h较 2 4h的菌落大 ,易于辨认 ;不同菌种在不同琼脂培养基上对嗜血杆菌琼脂卫星试验的结果有所不同。结论 卫星试验应在CO2 环境中进行 ,用脑心琼脂平板测试嗜血杆菌对X、V因子的需求结果较好 。 Objective To study which bacteria can be used in the satellite test for the identification of Haemophilus . Methods To inoculate Haemophilus on five types of media, to compare the X,V factor requirement test of Haemophilus with influence of cultivation time and the carbon dioxide (CO 2), to compare satellite test of different bacteria on five types of media. To compare various kinds of bacteria on various agar culture media, which influence the result of Haemophilus agar satellite test. Results The positive rate of X,V factor requirement on brain cardio agar (BCA) was higher than that of the nutrient agar and the MH agar ( P <0.01). The positive rate of X,V factor requirement and hemolytic test on 8%CO 2 was higher than that of the 0%CO 2. The positive rate of X,V factor requirement and hemolytic test on 48 h was higher than that on 24 h. The results of satellite test of sixty strains of Haemophilus were all positive. The results of BCA satellite test were better than that of the nutrient agar, and the results of the nutrient agar satellite test were better than the results of the M H agar; 48 hours satellite phenomenon was more obvious. The satellite phenomenon of most of strains which grew well on BCA, the nutrient agar and the MH agar was positive. Conclusions BCA is a better medium for X,V factor requirement of Haemophilus . We can use clinical separate strains of Staphylococcus or gram negative bacillus can be used for satellite test.
