摘要
目的 从基因水平探讨TNF α和TNF αR1在增生性瘢痕组织中的作用 ,为增生性瘢痕的基因治疗探索一条有效途径。方法 以正常瘢痕为对照 ,利用RT PCR技术分别检测 1年以上增生性瘢痕组织中成纤维细胞TNF α及TNF αR1mRNA的表达情况。结果 在增生性瘢痕组织中 ,TNF αmRNA、TNF αR1mRNA表达的相对值分别为 ( 1.2 8± 0 .5 5 )和 ( 1.0 2± 0 .13 ) ,均明显低于正常瘢痕组织 (P <0 .0 5 )。结论 提示TNF α在正常伤口的愈合过程中起着重要的作用 ,增生性瘢痕的形成可能与组织中TNF α和TNF αR1表达低下有关。基因治疗可提高早期增生性瘢痕中TNF α的含量或靶细胞膜上TNF αR1的数量与活性 。
Objective To explore the role of tumor necrosis factor alpha (TNF α) and its receptor (TNF α R 1) in hypertrophic scar at the gene level. Methods The expression of TNF α mRNA and TNF α R 1 in hypertrophic scars was tested by semiquantitative reverse polymerase chain reation (RT PCR) method and was compared with that in the normal scars. Results The expression ratios of TNF α mRNA and TNF α R 1 in hypertrophic scars were (1.28±0.55) and (1.02±0.13), respectively,which were significantly lower than those in the normal ones. Conclusion The results suggested that TNF α might play an important role in the process of wound healing and that the formation of hypertrophic scar might be due to the lack of TNF α or the down regulation of TNF α R 1. Therefore, it was reasonable to increase the level of TNF α or up regulate the amount of TNF α R 1 by using gene therapy in the early stage of hyphertrophic scar development.
