摘要
目的 探讨黏附分子ICAM 1、P selectin、E selectin、L selectin、PECAM 1和VCAM 1在肝脏移植中的表达及意义。方法 用免疫组织化学方法和原位杂交法 ,检测大鼠肝脏移植后不同时间 (1、3、5、7d)、不同模型中ICAM 1、P selectin、L selectin、E selectin蛋白、VCAM 1mRNA、PECAM 1mRNA的表达情况。结果 肝急性排斥组与自发耐受组相比 ,黏附分子ICAM 1、VCAM 1、PECAM 1、E selectin表达高 ;子代组与肝急性排斥组各指标表达类似 ,而ICAM 1的表达高于肝急性排斥组 ;半肝组与自发耐受组各指标表达类似 ,但ICAM 1、VCAM 1表达水平较自发耐受组高。P selectin、L selectin表达变化不明显。而正常大鼠肝脏未见黏附分子的表达。结论 肝排斥反应可能与黏附分子ICAM 1、VCAM 1、PECAM 1、E
Objective To investigate the effects of adhesion molecules including ICAM-1, VCAM-1, PECAM-1, P-selectin, L-selectin and E-selectin in rat liver allograft immune response in different stages. Methods After different rat allograft models were established in rats, ICAM-1, P-selectin, L-selectin and E-selectin were determined with immunohistochemistry. The expression of VCAM-1 mRNA and PECAM-1 mRNA was detected by in situ hybridization. Results Compared with liver allograft tolerance groups, the expression of adhesion molecules including ICAM-1, VCAM-1, PECAM-1 and E-selectin was enhanced in the acute liver allograft rejection group. The expression of ICAM-1 in F1 (P/D) D groups was higher than that in acute liver allograft rejection group. The expression of adhesion molecules detected in HLTx (P/2 D) group was similar with that in liver allograft tolerance group. There was no expression of the molecules in the normal rat liver. Conclusions Adhesion molecules including ICAM-1, VCAM-1, PECAM-1 and E-selectin may be involved in liver rejection response.
