摘要
目的 探讨失血性休克大鼠心肌细胞线粒体呼吸链复合体Ⅰ亚单位ND1、ND2编码基因改变。方法 通过建立失血性休克大鼠模型 ,提取心肌细胞线粒体DNA ,经PCR扩增呼吸链复合体Ⅰ亚单位ND1、ND2基因后测序检测其序列组成改变。结果 测序结果显示 ,失血性休克大鼠心肌细胞线粒体的ND1和ND2基因碱基序列无缺失、突变等改变 ;本实验中使用的Wistar大鼠ND1、ND2基因可能与NCBI提供的基因序列存在多个位点的单核苷酸多态性现象。结论 心肌细胞ND1。
Objective To observe the changes on gene sequence of the subunits ND1 and ND2 of mitochondrial respiratory complexⅠin rat heart cells after hemorrhagic shock.Methods The mtDNA(mitochondrial DNA) of heart cells in rat was extracted after hemorrhagic shock,and PCR(polymerase chain reaction) products of ND1,ND2 gene were sequenced.Results No deletions or mutations were found in the ND1 and ND2 genes of rat heart cells after hemorrhagic shock;on the other hand,multiple locus of single nuclear polymorphism in ND1 and ND2 genes may exist between the rats we used and that provided by NCBI.Conclusions The basepairs of ND1 and ND2 gene remain stable in the damage of hypoxia and ischemia after hemorrhagic shock in rat heart cells.
