生姜泻心汤防治大鼠反流性食管炎的作用机制探讨 被引量:10

Investigation of the Mechanisms of the Effects of Shengjiangxiexin Decoction in Preventing and Treating Reflux Esophagitis

摘要 目的 探讨生姜泻心汤防治大鼠反流性食管炎的作用机制。方法 大鼠 36只随机分为对照组、模型组、治疗组。制备反流性食管炎模型 ,对照组行伪手术。 1周后治疗组生姜泻心汤灌胃给药 ,其余两组予以生理盐水灌胃。术后 4周断头处死 ,食管作大体评分 ,并做组织病理学观察 ;制备食管组织匀浆 ,测定组织和血中的血管活性肠肽 (VIP)水平 ,并测定组织中丙二醛 (MDA)的含量及超氧化物歧化酶 (SOD)、谷胱甘肽过氧化物酶 (GSH PX)的活性。结果 生姜泻心汤能显著降低组织及血中VIP水平 ,降低组织中MDA的含量 ,提高SOD和GSH PX的活性 ,改进食管炎的大体和组织病理评分。结论 生姜泻心汤能降低大鼠反流性食管炎模型VIP水平、提高抗氧化能力 ,具有防治大鼠反流性食管炎的作用。 Objective To investigate the possible mechanisms of the effects of Shengjiangxiexin Decoction (SD) in preventing and treating reflux esophagitis. Methods 36 Rats were randomly divided into the control group, model group, and treating group; the operation for inducing reflux esophagitis was performed on the rats in the model group, and the sham-operation was performed on the rats in the control group. 1 Week after the operations, the treating group was given SD intragastrically, and the other two groups were given normal saline intragastrically. The rats in all groups were decollated 4 weeks after the operations, and their esophagi were collected for gross scoring and histopathologic observation and at the same time, the esophageal homogenate was prepared for measuring the levels of VIP in the esophageal tissue and the blood, the level of MDA in the esophageal tissue, and the activities of SOD and GSH-PX in the esophageal tissue. Results SD could markedly decrease the levels of VIP and MAD, and increase the activities of SOD and GSH-PX, and improve the results of gross scoring and with the histopathologic scale. Conclusion SD can decrease the level of VIP and increase the ability of antioxidation in the rat model of reflux esophagitis, and possesses the effects of preventing and treating reflux esophagitis.
