固相萃取-高效液相色谱法测定儿泻止颗粒中葛根素含量

Determination of puerain in Erxiezhi granules by SPE-HPLC method

目的 :本文采用固相萃取 -高效液相法测定了卫生部药品标准收载的儿泻止颗粒中葛根素含量。方法 :固相萃取条件 :C18小柱 ,30 %甲醇 2 0mL洗脱 ;色谱条件 :用十八烷基硅烷键合硅胶为填充剂 ;乙腈 水梯度洗脱 ;流速 1.0mL·min-1;检测波长 2 5 0nm。结果 :葛根素在 0 .0 0 6 2 2 8～ 0 .12 4 5 6 g·L-1范围内呈良好线性关系 (r =0 .9999) ,重现性RSD为 1.0 % ,n =5 ,回收率为 99.0 % ,RSD为 1.6 % ,n =5。结论 :本方法简便、准确且重复性好 ,可以有效控制该试剂中葛根素含量。

OBJECTIVE To establish a method for determinatio n of puerain in Erxiezhi granules by using SPE-HPLC. METHODS SPE condition: C 18 Cartridge. 20 mL 30% MeOH was used as elution solvent. HPLC condition: ODS column, a mixture of water and acetonit rile from (10∶90) to (40∶60) as the mobile phase. RESULTS Puerain had a good linearity in the rang of 0.006228 ～ 0.12456 g·L -1, recovery rate was 98.96% (RSD= 1.6%,n=5 ). CONCLUSION This method is simple, accurate, rapid and reproduci ble.
