摘要
为探讨家蚕外源基因导入法,开辟育种新途径,对家蚕精母细胞融合技术进行了初步研究,结果表明:(1)获取大量精母细胞的精巢摘出最适时期是四龄第2、3日。(2)精母细胞最适的生理盐水浓度是1.3%—1.4%。(3)利用微型振荡器的机械分离法,能有效地将精母细胞分离,其最适作用时间为10秒胶原酶对精母细胞的分离无论皮囊存在与否均无作用。(4)利用生理盐水的低渗处理可使邻接的精母细胞融合,但不能使完全分离的精母细胞融合。(5)聚乙二醇处理可使完全分离的精母细胞融合。
In order to approach the introduction of external genes (Bombyx mori)and to find new ways of breeding ,primary studies on the cell fusion of the spermatocytes have been carried out. The results were as in the following: (l)The 2nd or 3rd day after 3rd larval ecdysis is the optimal time for obtaining a large amount of spermatocytes from the testis. (2)The concentration of 1. 3 - 1. 4 percent physiological saline is the best for the spermatocytes. (3)The spermatocytes can be separated effectively by mechanic method of using mini-oscillator. The optimal treating time is 10s,and the collagenase is of no use on the separation of the spermatocytes whether there is cellulosa or not. (4)With the treatment of physiologiclal saline, the spermatocytes which linked to each other can fuse,but the entirely separated spermatocytes can not fuse. (5)The fully separated spen-matocytes can fuse by treatment with PEG.
