摘要
目的 建立钝挫伤白内障大鼠模型及探讨钝挫伤白内障与其晶状体上皮细胞凋亡的关系。方法 5 0只SD大鼠分成A、B、C、D、E共 5组 :分别用不同重量的小球 ,以不同次数连续从不同高度落下打击大鼠的左眼。裂隙灯下观察大鼠晶状体的变化 ,用透射电镜观察晶状体上皮细胞凋亡的超微结构 ,并与自身对照眼比较。结果 实验开始 1周后 ,5组实验眼的晶状体即出现不同程度的混浊 ,但是只有D组出现典型的、持续的环状混浊 ,其晶状体上皮细胞出现明显细胞凋亡的超微结构改变。各组自身对照眼晶状体上皮细胞超微结构无明显改变。结论 2 0cm高度 2 0g重量的外力持续打击可以使大鼠较快产生典型的、持续的白内障。钝挫伤白内障的发生与晶状体上皮细胞凋亡有关。
Objective To build contusion cataract model of rats and observe the relationship between contusion cataract and lens epithelial apoptosis. Methods Fifty 4-week-old SD rats were assigned to five groups according to the injury way.The left eyes of the rats were hit with a 10 g ball from 10 cm height continuously 100 times for group A,and from 20 cm height for group B,with a 20 g ball from 20 cm height 100 times for group C,continuously 100 times per week for five weeks totally in group D and 30 g ball from 20 cm height continuously 100 times per week for five weeks totally in group E.Changes of the lens of rats were observed after injury using slit-lamp microscope.After the lens opacification occurred,transmission electron microscope (TEM) was used to observe the ultrastructural changes of the lens epithelial cells.The fellow eyes of the rats were served as self-control. Results After one week,different changes appeared in the experimental eyes of the five groups.Thirty days later,the lens changes disappeared in group A,but the opacification was still existed in group B and C,and a typical vossius opacification only in D group. In the group E,contusion-induced other diseases and the morphological changes of apoptotic lens epithelial cells could be noticed in the experimental eyes.No obvious changes were found in the self-control eyes. Conclusion Typical and continuous cataract can be quickly induced by continuously hitting with 20 g weight foreign body from 20 cm height.Lens epithelial cells apoptosis is related with contusion cataract formation.
