摘要
目的 研究非霍奇金淋巴瘤(NHL)Survivin、p53的表达,探讨其在NHL发生、发展中的作用及相互关系。方法 应用免疫组织化学方法对54例NHL及10例反应性增生淋巴结石蜡包埋组织中Survivin、p53蛋白表达产物进行检测。结果 NHL组织中Survivin及p53蛋白的阳性率为51.9%(28/54)、40.7%(22/54)。随着NHL恶性程度的逐增,Survivin蛋白的表达强度及其阳性细胞密度逐渐增高,Survivin在低度恶性组NHL中的表达低于中-高度恶性组,分别为19%(4/21)、72.7%(24/33),两组之间差异显著(P<0.001)。p53-positive组Survivin表达显著高于p53-negative组,Survivin与p53表达之间显著相关(P<0.05)。结论 Survivi表达异常对促进NHL恶性增殖有一定作用。在NHL发病中Survivin与p53蛋白异常表达密切相关相关。分子水平干预Survivin蛋白表达可能提高淋巴瘤放、化疗疗效。
Objective:To identify the role of survivin, a novel inhibitor of apoptosis, in non-Hodgkin's lymphoma (NHL) and its relationship with expression of p53. Methods: Immunohistochemistry Envision system for survivin and p53 was conducted on 54 NHL and 10 reactive hyperplasia of lymph node (LN RH). Samples were archival paraffin-embedded blocks.Results: Survivin was detected in the cytoplasm of carcinoma cells in 51.9% (28/54) of NHL. p53 were detected in 40.7% (22/54) of NHL.Overexpression of survivin was related to the tumor grade. The number and immunoreactivity of survivin-positive cells in NHL significantly increased from low grade to high grade and they were 19% ( 4/21), 72.7%(24/33). There was statistically significance difference between two groups ( P <0.001). The positive expression rate of survivin (81.6 %) in the p53 positive group was higher than that in the p53 negative group (18.4 %) ( P <0.05). Expression of survivin and p53 was found to be significantly related. Conclusions:These data identify a role for survivin promoting aberrantly increased cell viability in NHL and suggest a potential correlation between accumulated p53 and survivin expression in neoplasia. Molecular manipulation of survivin expression may enhance chemotherapy and radiation therapy for NHL.
