摘要
目的 研究不同日龄胎鼠肺血管基膜IV型胶原及层黏连蛋白的发育情况 ,以探讨早产鼠易发生肺出血的原因。方法 分别用免疫组化法及原位杂交法检测日龄 17、18、19、2 1d胎鼠肺血管基膜IV型胶原、层黏连蛋白蛋白水平及mRNA水平的差异。结果 ① 17d胎鼠肺血管基膜IV型胶原及层黏连蛋白阳性产物的平均面积明显小于 2 1d胎鼠 ;② 17d胎鼠肺血管内皮细胞中IV型胶原mRNA和层黏连蛋白mRNA的表达明显弱于 2 1d胎鼠。结论 早产胎鼠肺血管基膜IV型胶原及层黏连蛋白的发育明显不成熟 ,且IV型胶原和层黏连蛋白在蛋白水平的表达受mRNA的调控。
Objective To investigate the causes of pulmonary hemorrhage which happened more in prexmature rats. We did research on the development of lung vessel basement membrane of collagen IV(CoIV) and lamina(lam)with different gestation fetal rats.Methods The protein and mRNA of CoIV and lam of lung vessels are measured by immunohistochemical technology and in situ hybridization of Wistar rats with gestational days for 17, 18, 19 and 21. Results ①The average area of CoIV and lam of 17 days group is much less than that of 21 days group. ②The mRNA expression of CoIV and lam in lung endothelial cell of 17 days group is much less than that of 21 days group.Conclusion The development of CoIV and lam of lung vessel basement membrane are obviously premature in premature rats and the expression of protein level of them are modulated by mRNA, which is one of causes for pulmonary hemorrhage in prematures.
