摘要 目的 采用反相离子对HPLC法测定人血浆中苯溴马龙的浓度。方法 血浆样品中苯溴马龙及内标美洛昔康经氯仿提取 ,在SymmetryC18柱上 ,以乙腈 - 0 .0 1mol·L-1NH4H2 PO4(含 0 .5mmol·L-1四辛基溴化铵 ) (73∶2 7)为流动相 ,于 35 0nm检测。结果 标准曲线线性范围 0 .0 312 5~ 4 .0mg·L-1,萃取回收率 81.9%~ 89.5 % ,加样回收率 99.7%~ 10 0 .3% ,日内测定RSD为 1.4 1%~ 2 .0 9% (n =7) ,日间测定RSD为 2 .5 9%~ 5 .16 % (n =7)。结论 所用方法简便、准确 ,重复性好 ,用于苯溴马龙在人体药动学研究中的血药浓度测定结果良好。 OBJECTIVE To develop a method for the determination of Benzbromarone in human plasma by ion-pair HPLC.METHODS Meloxicam was used as the internal standard,After the plasma sample was extracted with 2 ml chloroform,separation was achieved on the Symmetry C 18 (150 mm×3.9 mm, 5 μm) column.The mobile phase was consisted of CH 3CN - 0.01 mol·L -1 (NH 4)H 2PO 4(containing 0.5 mmol·L -1 tetraoctylammonium bromide)(73∶27)and the effluent was measured by UV of the wavelength of 350 nm.RESULTS The calibration curves were linear within the range of 0.03125 to 4.0mg·L -1.The limit of detection was 0.01 mg·L -1.The extraction recovery of Benzbromarone were 81.9% to 89.5%.The methodological recovery were 99.7% to 100.3%.The within-day RSD was 1.41% to 2.09%,and between days RSD was 2.59% to 5.16%.CONCLUSION The assay was simple,rapid,and sensitive.It is satisfied to be used to study the pharmacokinetics and relative bioavailability of Benzbromarone.
