人Tamm-Horsfall蛋白(THP)自身免疫抗体快速AB-ELISA法的建立

The Establishment of Rapid Analysis of Human Self-antibody of Tamm-Horsfall Protein by Anti-Biotin Enzyme-Linked Immunosordent Assay(AB-ELISA)

目的 建立人 Tamm- Horsfall蛋白 ( THP)自身免疫抗体的快速 AB- EL ISA测定法。方法 在提取纯化人 Tamm- Horsfall蛋白 ( THP)后 ,制备生物素 - THP,建立了人 THP自身免疫抗体 ( Ig G)快速 AB-ELISA(夹心 )测定法。结果 此法标准曲线范围 1～ 5 0μg/ml,灵敏度 1μg/ml,实验内变异系数 6.5 6% ,实验间变异系数 1 0 .35 % ,同时测定 1 0 0例正常人 THP自身免疫抗体 ( Ig G)为 0～ 33.81 μg/ml。结论 经实验表明该法灵敏 ,重复性较好 。

Objective To establish the rapid analysis of human self-antibody of Tamm-Horsfall protein(THP) by anti-biotin enzyme-linked immunosordent assay(AB-ELISA).Methods Tamm-Horsfall protein was extracted and purified from urine.Synthesis THP with Biotin labelling,to establish the rapid analysis of human self-antibody (IgG) of Tamm-Horsfall protein(THP) by anti-biotin enzyme-linked immunosordent assay(AB-ELISA).Results The standard curve range of this methods was 1～50 μg/ml,the sensitivity was 1 μg/ml.The variance within batch and between batch were 6.56% and 10.35%.The human self-antiboby (IgG) of Tamm-Horsfall protein of 100 healthy was 0～33.81 μg/ml.Conclusion This method was sensitive and with a good repeatability.So it's a good method for clinical laboratory assay.
