摘要
采用异硫氰酸胍和Catrimox 14TM联合变性,酚、氯仿和异戊醇抽提的方法,对真菌总RNA进行提取。得到的RNA样品经过紫外分光光度法和甲醛变性凝胶电泳检测,证实为完整、均一的总RNA,为构建真菌的cDNA文库和筛选差异表达的基因奠定了基础。
The method was established for RNA isolation from fungi using Guanidinium isothiocyante and Catrimox-14^(TM) for denaturing, and water-satured phenol chloroform and isoamylalcohole for extracting. The integrity and purification of the total RNA isolated are eligible by using formaldehyde-a-garose gel electrophoresis and UV spectrophotometer。 It is useful in constructing cDNA library for the total RNA and also valuable in screening differential expressed genes.
