大鼠脑局灶缺血/再灌注对外周血单个核CD34^+细胞的影响

摘要 目的 :本研究探讨大鼠脑局灶缺血 /再灌注是否影响外周血单个核CD3 4+ 细胞的数量。方法 :采用大鼠大脑中动脉 (MCA)缺血 /再灌注线栓法模型 ,将大鼠随机分为 3组 :对照组 ,假手术组 ,动物模型组。取再灌注后 1、3、6、12h和 1、2、3、4、7、14d ,10个观察点 ,测外周血单个核CD3 4+ 表达情况。结果 :大鼠脑缺血 /再灌注模型中外周血单个核CD3 4+ 细胞在再灌注后 1h~ 2d无明显变化 ,在脑缺血 /再灌注后 3~ 7d明显减少 ,14d后恢复。结论 :大鼠脑缺血 /再灌注模型再灌注后 1h~ 2d再灌注后外周血单个核CD3 4+ 细胞没有变化 ,在脑缺血 /再灌注后 3~ 7d明显减少。 Aim:This study was intended to detect changes of circulating CD34 + cells after focal cerebral ischemia/reperfusion in rats.Methods:A model of a suture occlusion of right middle cerebral artery (MCA) of male SD ratd was used.60 mature male SD rats were randomly divided into 12 groups:control group,sham-operated control group,1-hour (1h),3h,6h,12h,24h,2-day (2d),3d,4d,7d,,14d reperfusion groups.After right middle cerebral artery occlude,the rats were suffered from ischemia for 2 hours, then reperfusion was allowed for different time as mentioned above before decapitation.Circulating CD34 + cells were detected by cytoflowmetric analysis.Results:There was no influence on circulating CD34 + cells in rat MCAO models from 1h to 2 days after reperfusion.But circulating CD34 + cells sharply decreased from 3 to 7 days after reperfusion and returned to basal values at 14th day after cerebral reperfusion.Conclusion:Circulating CD34 + cells sharply decreased from 3 to 7 days after reperfusion in rat MCAO models.
