摘要
目的:探讨在纤细眼虫(Euglenagracilis)中是否存在核纤层蛋白(lamin)基因,为核纤层蛋白基因的起源与进化研究提供线索.方法:以较为低等生物的核纤层蛋白基因cDNA序列为参考,设计引物P0010和P0012,以纤细眼虫总DNA为模板进行PCR,PCR产物回收、测序.由于不能获取完整序列,所以据测序结果又设计了P0013和P0014两条中间引物,组成P0010/P0013和P0014/P0012引物对进行PCR,回收产物测序.结果:P0010/P0012、P0010/P0013和P0014/P0012引物对进行PCR分别获得775、400和395bp的特异性PCR产物,对这3种产物分别测序,最终获得了P0010和P0012之间的全序列,共775bp.结论:在纤细眼虫中存在着核纤层蛋白基因.
Aim:To investigate whether Euglena gracilis possesses lamin gene. Methods: According to the conservative domains of some known lamin cDNAs of primitive metazoa, a pair of primers (P0010, P0012) were designed to amplify the tail domain of lamin gene. From total DNA of Euglena gracilis, a fragment was amplified and sequenced. But it was not able to get the whole sequence of the PCR product, so according to the sequences having been sequenced, P0013 and P0014 which were respectively in paires with P0010 and P0012, were designed to PCR. Then the PCR products were sequenced. Results: Three specific PCR products of 775 bp, 400 bp and 395 bp were amplified, which respectively corresponded with the primer pairs of P0010/P0012, P0010/P0013 and P0014/P0012. These PCR products were sequenced, then the sequences between P0010 and P0012 was obtained. Conclution: Our study suggests that Euglena gracilis possesses lamin gene.
