脑胶质瘤的恶性增殖与DNA拓扑异构酶Ⅱα的表达研究

A study on relationship between expression of topoisomeraseⅡa lpha and malignant proliferation of glioma

目的 通过观察DNA拓扑异构酶Ⅱα(TopoⅡα)在不同级别脑胶质瘤中表达 ,研究TopoⅡα表达与脑胶质瘤分化程度的关系。方法 应用免疫组化二步法对 5 9例脑胶质瘤标本及 5例正常脑组织中TopoⅡα的表达进行对照研究。结果 随着脑胶质瘤病理级别的升高 ,TopoⅡα标记指数也升高 ,低度恶性组与中、高度恶性组TopoⅡα表达差异均有显著性 (P <0 .0 5 ) ,而中度恶性和高度恶性组间差异无显著性 (P >0 .0 5 )。结论 TopoⅡα能较准确地反映脑胶质瘤增殖潜能和恶性程度 。

Objective To observe the exp ression of DNA topoisomeraseⅡalpha (TopoⅡα)in gliom a of various pathological grades and to study the relationship between TopoⅡα a nd tumor grades.Methods Two-step immunohist ochemical staining technique was used to study the expression of TopoⅡα in 59 cases of gliomas of various pathological grades,and in 5 cases of brain tissue as the normal control. Results TopoⅡα positive labeling index increased with increase in the pathological grade of glioma,with significant diffe rence between low and middle,high malig nant glioma groups(P<0.05),while the difference between middle and high malignant glioma groups was not statistically significant(P>0.05). Conclusion TopoⅡ α detection may accurately reflect the malignant degree and potential cell p r oliferation activity of glioma,and provide an important reference for estimating the prognosis of the patients with glioma.
