摘要
目的:本实验目的是研究虫草素(Cordycepin)对HBSS平衡盐溶液诱导的原代培养的大鼠肾小球系膜细胞(glomerular mesangial cells, GMCs)自噬的保护作用。方法:大鼠GMCs分离后分别在含有细胞培养基、高糖溶液、HBSS溶液、HBSS溶液 + 虫草素培养基中培养,培养24 h、72 h后用Western blotting分析方法检测细胞自噬标记LC3 I、II和p62/SQSTM1。结果:培养24 h,各组之间LC3和p62/SQSTM1表达没有显著差异,72 h后在高糖培养基中的细胞LC3-I、LC3-II、p62/SQSTM1下调,在含HBSS溶液培养基中的细胞LC3-I、LC3-II、p62/SQSTM1表达均上调,在含HBSS溶液 + 虫草素培养基中的细胞LC3-I、LC3-II、p62/SQSTM1则相反,表达均下降。结论:虫草素可以抑制HBSS溶液诱导的肾小球系膜细胞的自噬作用。
Objective: The aim of this study was to investigate the effects of cordycepin on the impairment of autophagy induced by HBSS in primary cultured rat glomerular mesangial cells (GMCs). Methods: Rat GMCs were isolated and cultured in cell medium, cell medium + high-glucose, HBSS and HBSS + cordycepin medium. At 24 h and 72 h of culture, the cells were examined for expression levels of the autophagy markers LC3 I, II and p62/sequestosome-1 (SQSTM1) using western blotting analysis. Results: At 24 h, no significant difference in the expression of LC3 I, II and p62/SQSTM1 was observed among the groups;however, at 72 h the cells exposed to high-glucose medium showed down-regulated LC3 I, II and p62/SQSTM1 expression. The cells exposed to HBSS for 72 h showed up-regulated LC3 I, II and p62/SQSTM1 expression. These changes were reversed in the HBSS + cordycepin group at 72 h. Conclusions: In conclusion, Cordycepin can inhibit HBSS-induced autophagy in rat GMCs.
