心脏重编程在心肌梗死修复中的研究进展

摘要 人类成年心脏缺乏强大的内源性修复机制来完全恢复损伤后的心功能,因此,再生和修复受损心肌的能力是治疗心力衰竭或心肌梗死的首要任务。在体外和体内直接将成纤维细胞转化为心肌样细胞为这一问题提供了巨大的希望。此外,心肌成纤维细胞的直接重编程可规避干细胞疗法在临床应用中产生的许多问题,且为心脏再生和修复提供了有力的治疗策略。在这篇综述中,我们阐述了心脏直接重编程研究的进展,以及改善心脏重编程的效率的方法。最后,我们讨论了未来临床应用的更安全的重编程方法的发展。 The human adult heart lacks a strong endogenous repair mechanism to completely restore the in-jured heart function. Therefore, the ability to regenerate and repair damaged myocardium is the primary task in the treatment of heart failure or myocardial infarction. Direct transformation of fi-broblasts into cardiomyocytes in vitro and in vivo provides great hope for this problem. In addition, direct reprogramming of cardiac fibroblasts can avoid many problems in clinical application of stem cell therapy, and provide a powerful therapeutic strategy for heart regeneration and repair. In this review, we describe the research progress of direct cardiac reprogramming and the methods to im-prove the efficiency of cardiac reprogramming. Finally, we discuss the development of safer repro-gramming methods for clinical application in the future.
