摘要
研究了高压脉冲等离子体射流(APPJ)对耐药性鲍曼不动杆菌(A. baumannii)生物膜的失活效果和机制。研究发现,该脉冲等离子体射流对生物膜中的细菌有明显的灭活作用。菌落形成单位(CFU)计数方法处理显示,30分钟处理后,生物膜中99.9%的细菌失去其可培养发育能力。与此同时,刃天青荧光染色方法显示约80%的细菌失去了新陈代谢能力,部分细菌进入了一种存活但不可培养发育的状态(VBNC),H2-DCFDA荧光染色方法显示,等离子体处理后生物膜中细菌内的活性含氧基团(ROS)的浓度有所提高。等离子体活性物种可能诱导了细菌内部应激反应的产生,导致胞内ROS的生成,最终氧化破坏细菌结构,这可能是等离子体导致生物膜中细菌失活的重要原因。
The inactivation effect and mechanismof high atmospheric pressure pulsed plasma jet(APPJ)on drug-resistant Acinetobacter baumannii(A.baumannii)Biofilm were studied inthis paper.It is found that the pulsed plasma has a remarkable ability toinactivate bacteria in biofilm.The CFU countingmethod of colony forming units showed that almost all the bacteria(99.9%)inthe biofilm lost their culturability after 30 minutes of treatment.At the sametime,about 80%of the bacteria had lost their metabolism,and some of thebacteria had entered a viable but uncultured state of VBNC.H2-DCFDA fluorescence staining showed that theconcentration of active oxygen-containing group(ROS)in biofilm was increasedafter plasma treatment.Plasma active species may induce internal stressreaction of bacteria,lead to a large number of intracellular ROS production,and ultimately oxidize the bacterial structure,which may be an importantreason for the inactivation of bacteria in biofilm caused by plasma.
