摘要
外泌体具有多种生物学功能,对疾病的治疗和诊断具有重要的意义,而获得高纯度的外泌体对后续的研究至关重要。本实验采用溶剂热法,以钛酸四异丙酯作为前驱物,EDTA作为包裹剂,优化实验条件,制备出粒径为1~3 μm、分散性良好、表面光滑的TiO2微球。利用TiO2微球表面Lewis酸位点实现对外泌体的特性吸附及纯化,并采用扫描电镜(SEM)、透射电镜(TEM)进行表征观察。结果表明,TiO2微球能够高效地吸附外泌体,使用10%氨水可以快速将外泌体从TiO2微球表面洗脱下来且不破坏外泌体的囊泡结构。
Exosome has a variety of biological functions and is of great significance for the treatment and di-agnosis of diseases. And it is very important to obtain high purity exosome for subsequent research. In this experiment, TiO2 microspheres were prepared by hot solvent method with tetrai-sopropyltitanate as precursor and EDTA as precipitant. Optimizing experimental conditions, TiO2 microspheres with particle size of 1~3 μm, good dispersion and smooth surface were successfully prepared. Exosomes were adsorbed and purified by Lewis acid sites on the surface of TiO2 micro-spheres, and characterized by scanning electron microscopy (SEM) and transmission electron mi-croscopy (TEM). The results show that TiO2 microspheres could adsorb efficiently exosome. 10% ammonia could quickly elute exosome from the surface of TiO2 microspheres without destroying the vesicle structure of exocrine.
