摘要
目的:探讨愈疡胶囊对铜绿假单胞菌(P. aeruginosa)的抑菌作用及对大鼠烫伤感染的治疗。方法:采用杯碟法研究愈疡胶囊对P. aeruginosa的体外抑菌效果,扫描电镜观察细菌膜的受损情况。将50只SD大鼠随机划分为5组:模型对照组,愈疡胶囊高、中、低剂量组(157.5, 315, 630 mg/kg)和阿莫西林(125 mg/kg)阳性对照组,每组各10只。将大鼠背部烫伤并涂抹菌液建立大鼠烫伤感染模型,采用灌胃方式每天给药一次,持续8天。记录大鼠一般体征如积脓、红肿、溃疡等并分级评分。取感染部位皮肤组织,匀浆铺板统计菌落数、测量炎症因子含量,并进行H&E染色观察组织恢复的病理学过程。结果:杯碟法实验表明高剂量组抑菌圈直径(1.3992 ±0.0080 cm)明显大于对照组(0.6 ±0.0086 cm),差异有显著性(P 【0.01)。扫描电镜结果显示愈疡胶囊治疗后可导致P. aeruginosa细菌膜出现皱褶、凹陷和变形,进而杀灭细菌。体内动物实验结果表明愈疡胶囊能够明显减少感染区域的细菌定植数量,高剂量组菌落数(117 CFU)明显低于模型对照组(694 CFU) (P 【0.01);通过调节炎症因子水平促进伤口愈合;H&E染色结果和伤口愈合速率证明愈疡胶囊能够促进大鼠伤口愈合,减缓组织炎症,促进粒状组织形成,有利于表皮新生。结论:愈疡胶囊对P. aeruginosa具有较好的抑菌作用,可用于临床治疗P. aeruginosa引起的烫伤感染。
Objective: To study the antibacterial effect of Yuyang capsule on P. aeruginosa and promoting repair on scalded infection of rats. Methods: The cup and saucer method was used to study the antibacterial effect of Yuyang Capsule against P. aeruginosa in vitro, and the damage of the bacterial membrane was observed by scanning electron microscope. SD rats were randomly divided into 5 groups: model control group, Yuyang capsule high, medium and low dose groups (157.5, 315, 630 mg/kg) and amoxicillin 125 mg/kg group (positive control group), 10 in each group. The rats were scalded on the back and smeared with bacterial liquid to establish a scalded infection model. The rats were given intragastrically once a day, for 8 days. The rats’ general physical signs such as empyema, redness and ulcers were recorded and scored. The number of colonies was counted by agarose plating. It promotes wound healing by regulating the levels of inflammatory cytokines and the skin tissue was stained with H&E to observe the pathological process of tissue recovery. Results: The cup and saucer method experiment showed that the diameter of the inhibition zone of the high-dose group (1.3992 ±0.0080 cm) was significantly larger than that of the control group (0.6 ±0.0086 cm, and the difference was significant (P <0.01). Scanning Electron Microscope results showed that Yuyang capsule could induce the bacterial membrane of P. aeruginosa to be wrinkled, dented and deformed, and eventually killed the bacteria. The results of in vivo animal experiments showed that Yuyang capsule can significantly reduce the number of bacterial colonization in the infected area. The number of colonies in the high-dose group (117 CFU) is significantly lower than Model control group (694 CFU) (P <0.01);H&E staining results proved that Yuyang Capsule can promote wound healing in rats, slow down tissue inflammation, promote granular tissue formation, and be beneficial to epidermal regeneration. Conclusion: Yuyang Capsule has an excellent antibacterial effect against P. aeruginosa and could be applied for treating the scald infection caused by P. aeruginosa.
