养阴扶正汤调节Lewis肺癌荷瘤鼠T细胞耗竭的实验研究

摘要 目的:探讨养阴扶正汤调控Lewis肺癌荷瘤鼠T细胞耗竭的机制。方法:应用免疫磁珠法筛选出PD-L1+Lewis肺癌细胞系,分别构建PD-L1+Lewis及Lewis肺癌荷瘤鼠模型。造模成功后,将PD-L1+Lewis及Lewis肺癌荷瘤鼠按随机数字表法分组,分为模型组、PD-L1抑制剂组(阿特珠单抗)、养阴扶正汤组、联合组(养阴扶正汤 + 阿特珠单抗)。药物干预10天后,利用ELISA检测各组脾脏淋巴细胞分泌IL-2、IFN-γ、IL-10、TGF-β的水平。结果:PD-L1+Lewis及Lewis肺癌荷瘤鼠养阴扶正汤治疗组脾淋巴细胞分泌IL-2、IFN-γ水平均增高(P 【0.05),分泌IL-10、TGF-β水平均降低(P 【0.05)。结论:养阴扶正汤能逆转T细胞耗竭状态,有望成为有效逆转PD-1+T细胞耗竭的中药方剂。 Objective: To explore the mechanism of Yangyin Fuzheng Decoction in regulating T cell exhaus-tion of mice with Lewis lung cancer cell. Methods: The cell lines of PD-L1+Lewis lung cancer were screened by immunomagnetic bead method, and the C57BL/6 mice models of PD-L1+Lewis and Lewis lung cancer cell were constructed respectively. After successful modeling, C57BL/6 mice with PD-L1+Lewis and Lewis lung cancer cell were divided into model group, PD-L1 inhibitor group (Atezolizumab), Yangyin Fuzheng Decoction group, and combined group (Yangyin Fu-zheng Decocotion + Atezolizumab) according to the random number table method. After 10 days of drug intervention, ELISA was used to detect the levels of IL-2, I IFN-γ, IL-10 and TGF-β secret-ed by spleen lymphocytes in each group.
