miRNA221对宫颈癌细胞株Siha侵袭转移行为的影响

Effect of miRNA221 on the invasion and metastasis of cervical cancer cell lines Siha

目的 :探讨干扰miRNA221对宫颈癌细胞侵袭转移行为的影响。方法 :实验分为Siha,Siha-模拟物即(siha-mimic),阴性对照Siha-nc(mimic),Siha-抑制剂即(Siha-inhibitor),阴性对照Siha-nc(inhibitor)五组。采用q PCR、Western blotting、划痕实验和Transwell小室实验对miRNA221在宫颈癌siha细胞株侵袭转移中的作用。结果:在siha-mimic组,miRNA221表达明显高于Siha组和Siha-nc(mimic)组,E-钙粘素(E-cadherin,E-cad)表达水平明显降低,N-钙粘素(N-cadherin,N-cad)及波形蛋白(Vimentin)蛋白表达水平明显上调;在Siha-inhibitor组,miRNA221表达明显低于Siha组和Siha-nv(inhibitor)组,E-cad蛋白表达水平明显升高,N-cad和Vimentin表达水平显著下调。结论 :干扰miRNA221抑制宫颈癌细胞侵袭转移。

Objective To investigate the effect of miRNA221 on the invasion and metastasis of cervical cancer cell lines Siha.Method The experiment was divided into five groups: Siha, Siha-nc (mimic), Siha-nc (inhibitor), Siha-mimic, Siha-inhibitor. Testing the effect of miRNA221 on the invasion and metastasis of cervical cancer cell lines Siha by qPCR, Western blotting, the scratch test and Transwell chamber experiment.Results In Siha-mimic group, the expression of miRNA221 was significantly higher than that in Siha group and Siha-nc (mimic) group, the expression level of E-cad significantly decreased, the protein expressionsof N-cad and Vimentin significantly increased in Siha-inhibitor group, the expression of miRNA221 was significantly lower than that in Siha group and Siha-nc (inhibitor) group, the expression level of E-cad protein significantly in-creased, the expression levels of Vimentin and N-cad significantly decreased.Conclusion miRNA221 inhibits the invasion and metastasis of cervical cancer cell lines Siha.
