摘要
为了获得樱桃谷鸭寡腺苷酸合成酶样蛋白质基因(OASL)全长cDNA,并且初步了解其生物学特性,采用RT-PCR和RACE方法从鸭脾脏组织总RNA中扩增OASL基因cDNA片段,应用生物信息学方法,系统分析鸭OASL的遗传进化以及蛋白质的理化性质、二级结构和亚细胞定位。结果表明,樱桃谷鸭OASL基因(Gen Bank登录号:KX255654)全长1 630 bp,其中包含19 bp的5'UTR(Untranslated region,UTR)、99 bp的3'UTR(Untranslated region,UTR)和PolyA尾巴、1 512 bp的编码区(Coding region,CDS),翻译编码504个氨基酸多肽。鸭OASL蛋白具有寡腺苷酸合成酶(OAS)蛋白家族的典型特征,即N端为寡腺苷酸合成酶样结构域(OAS-like domain,OLD),C端为2个串联的泛素化样结构域(Ubiquitin-like domains,Ub LDs),不具有信号肽和跨膜区。序列比对和系统进化分析结果表明,OASL氨基酸序列在同种之间比较保守,与已报道的鸭OASL的同源性高达100%,与鹅同源性为78%,与人、鼠和猪等哺乳动物的同源性仅为43%~45%。
To analyze the biological charateristics of oligoadenylate synthase-likely gene (OASL) in cherry valley duck, the complete cDNA of duck OASL gene was obtained by RT-PCR, 5′RACE and 3′RACE methods.The genetic evolutionary relationships, physical and chemical characteristics, second structure and subcellular localization of OASL were analyzed by bioinformatics tools.Results were as follows.The full-length cDNA sequence of OASL gene is 1 630 bp in size, including a 19-bp 5′ UTR, a 99-bp 3′UTR with the poly (A) tail (GenBank accession No.KX255654) and an open reading frame (ORF) of 1 512 bp in length, and encodes a predicted protein of 504 amino acids.OASL protein shared some typical characteristics of OAS family which is one OAS-like domain in the N-terminal and two tandem ubiquitin-like (UBL) domains in the C termined.No putative signal peptide and transmembrane domain were predicted.Sequence alignment and phylogenetic analysis showed that duck OASL was highly conserved among the same population, which shared 100% homology of aminoacid sequence with Anas platyrhynchos, 78% with Anser cygnoide.
