摘要
为了探讨梨凝集素受体蛋白激酶(LecRLK)对非生物胁迫(盐、渗透胁迫)的响应情况,以梨PcLRLK066基因为研究对象,研究LRK066在梨和其他物种间的进化关系及其在豆梨不同组织中的表达情况,并观察了其在细胞中的定位情况,此外还研究了转PcLRLK066拟南芥在不同盐浓度和甘露醇浓度处理下的发芽情况。结果显示,豆梨中PcLRLK066与苹果进化距离最近,与野生番茄最远;PcLRLK066在不同组织器官中表达差异显著;通过亚细胞定位发现PcLRLK066可能定位在细胞膜上;转入PcLRLK066基因后的拟南芥在盐和渗透胁迫处理下发芽率均低于野生型拟南芥,表明转基因拟南芥对盐和干旱更加敏感,PcLRLK066可能参与了盐胁迫和渗透胁迫。
To explore the role of pear LecRLK in response to abiotic stress such as salt and osmotic stress, the evolutionary relationship of PcLRLK066 in pear and other species were studied.The phylogenetic tree showed that PcLRLK066 in pear and apple had the nearest evolutionary distance to apple but the farthest to while wild tomato.PT-PCR revealed that PcLRLK066 were differentially expressed in different organs of pear.PcLRLK066 was subcellularly localized on plasma membrane.The germination rates of transgenic Arabidopsis plants under salt and osmotic stress were lower than those in wild type Arabidopsis plants, indicating that PcLRLK066-transgenic Arabidopsis is more sensitive to abiotic stress and PcLRLK066 might be involved in the responses.
