摘要
目的 探讨TPX2基因对肺癌细胞迁移侵袭及对基质金属蛋白酶2与基质金属蛋白酶9表达的影响.方法 将TPX2基因的重组载体p Magic4.1-shRNA-TPX2转入人肺腺癌A549细胞后,采用Transwell小室法检测细胞的迁移侵袭能力,逆转录-聚合酶链反应和蛋白质印迹法检测A549细胞中TPX2、基质金属蛋白酶2与基质金属蛋白酶9的表达水平.结果 实验组A549细胞的迁移侵袭能力均显著低于阴性对照组和空白对照组(P<0.05).实验组TPX2、基质金属蛋白酶2、基质金属蛋白酶9的mRNA与蛋白表达水平均显著低于阴性对照组与空白对照组(P<0.05或0.01);阴性对照组与空白对照组比较差异无统计学意义(P>0.05).结论 TPX2基因能够明显抑制肺癌细胞的侵袭和转移,靶向沉默TPX2基因可显著降低基质金属蛋白酶2、基质金属蛋白酶9基因的表达水平.
Objective To investigate the effects of TPX 2 gene on migration and invasion of lung cancer cells and expression of MMP-2 and MMP-9. Methods After transfection with the recombination TPX 2 gene-silencing p Magic4.l-sh RNA-TPX2,the abilities of proliferation and invasion of A549 cells were detected by Transwell chamber assay, respectively. The expression levels of TPX2, MMP-2,MMP-9 were measured by RT-PCR and Western blotting, respectively. Results The abilities of proliferation and invasion of A549 cells in the experimental group were significantly lower than those in the negative control group and the blank control group (P<0.05). The expression levels of TPX2, MMP-2, MMP-9 mRNA and protein in the experimental group were significantly lower than those in the negative control group and the blank control group (P<0.05 or 0.01); there was no statistical significance between the negative control group and the blank control group (P>0.05). Conclusion TPX2 gene can obviously inhibit the migration and invasion of lung carcinoma cells, silent TPX2 gene can significantly decrease the expressions of MMP-2, MMP-9 gene.
