MBL在绵羊呼吸道上皮细胞抗肺炎支原体感染过程中的机制探讨

摘要 为了探究甘露糖结合凝集素(MBL)对绵羊呼吸道黏膜上皮细胞抵御绵羊肺炎支原体(MO)感染的作用,本研究从绵羊血清中提取天然MBL蛋白,将绵羊呼吸道黏膜上皮细胞分4组,分别处理为:接种MO前细胞培养基中添加MBL蛋白;接种MO后细胞培养基中添加MBL蛋白;接种MO后继续细胞培养;正常培养。采用荧光定量PCR的方法,对各组细胞MBL、C3、ICAM-1、IL-6、SBD-1基因的表达量进行研究。结果显示:与对照组相比,其他组细胞密度均减少,且MBL、C3基因的表达量显著下降(P<0.05),同时ICAM-1、IL-6基因的表达量显著升高(P<0.05);接种MO前加入MBL的细胞ICAM-1基因的表达量相对其他组显著升高(P<0.05),且其SBD-1基因表达量极显著升高(P<0.01)。被MO感染的各组细胞均产生免疫应答,在感染前加入MBL组细胞的免疫应答更强烈,该过程或有ICAM-1和SBD-1参与。 To explore the function of mannose-binding lectin(MBL) during the process that sheep mucosal epithelial cells against Mycoplasma ovipneumoniae infection. Natural MBL was extracted from sheep serum. The sheep mucosal epithelial cells were divided into 4 groups. Operation as fallow: added MBL in cell culture before inoculation of MO; added MBL in cell culture after inoculation of MO; cultured cells after inoculation of MO; cultured normally. The genes expression levels of MBL,C3, ICAM-1, IL-6, SBD-1 was detected by fluorescence quantitative PCR after reverse transcription. The results showed that,compared with the control group, the density of other groups were decreased, and the expression of MBL and C3 gene was significantly decreased(P<0.05), while the expression of ICAM-1 and IL-6 gene was significantly increased(P<0.05); the expression of MBL+MO ICAM-1 gene relative to other groups increased significantly(P<0.05), and the expression of SBD-1 the amount of gene significantly increased(P<0.01). All the cells infected by MO were immune response, and the immune response of the group, MBL was added before infection, was more intense. The ICAM-1 and SBD-1 may be the participants during these process.
