SLC6A4基因表达及多态性对吐鲁番斗鸡攻击行为的影响

The influence of the expression and polymorphism of SLC6A4 gene on the aggressive behaviour of Turpan gamecock

为探讨SLC6A4基因对吐鲁番斗鸡攻击行为的调控作用,寻找吐鲁番斗鸡攻击行为的分子标记,本研究分析吐鲁番斗鸡、新罗曼蛋鸡及其杂交F1代、F2代脑组织初生到6月龄各月龄的SLC6A4基因表达水平,筛查亲代、F1代和F2代的SLC6A4基因SNP,结合攻击行为表型分析验证SLC6A4基因多态性与吐鲁番斗鸡攻击行为的关联性。结果显示:初生到6月龄的吐鲁番斗鸡和新罗曼蛋鸡脑组织的SLC6A4基因表达呈波浪形,与攻击行为呈显著的正相关;在SLC6A4基因第一外显子上发现了2个错义突变位点(T6213768G、C6213750G),而且BB基因型的攻击性显著高于其他基因型,因此,SLC6A4基因可作为吐鲁番斗鸡攻击行为选择的候选基因,建议把第一外显子的BB基因型确定为吐鲁番斗鸡攻击行为分子标记的参考基因型。

To explore the regulation function of SLC6A4 gene on the aggressive behavior of Turpan gamecock. The expression level of SLC6A4 gene in the brain of Turpan gamecock, Roman laying hens and their F1 population from newborn to6-momth-old were detected. The SNPs of SLC6A4 gene were detected by SSCP and sequencing method. The results showed that the expression level of SLC6A4 gene was wavy from born to 6-month-old and correlate positively with aggressive behavior.Two missense mutations in the first exon were found, and the aggressive scores of cocks with BB genotype were significantly higher than that of other genotypes and could be used as a marker of aggressive behavior. Results showed that the SLC6A4 gene from birth to 6-mon age of Turpan gamecock and New Roman laying hens brain tissue showed wavy expression. The aggressive behavior weas positive correlated with exon 1 of SLC6A4 gene.There were two base mutations(T6213768G,C6213750G) and lead to the change of the amino acids. SSCP analysis was found SLC6A4 gene exon 1 of BB genotype of aggression was significantly higher than other genotypes, which can be taken as reference to the Turpan gamecock molecular auxiliary marker selection genotype.
