摘要
目的 探讨一种改进的微栓子栓塞法构建大鼠脑缺血模型的效果.方法 将体重250~300 g的雄性SD大鼠30只随机分成假手术组、50个栓子数量组、100个栓子数量组,使用改进的微栓子栓塞法构建大鼠脑缺血模型,术后24 h进行神经行为学评分,随后处死,行TTC染色计算梗死体积.结果 假手术组神经行为学平均评分为0分,50个栓子数量组与100个栓子数量组神经行为学评分集中于2分,100个栓子数量组的平均评分要高于50个栓子数量组.假手术组的大鼠脑组织未出现梗死,而50个与100个栓子数量组的大鼠脑组织梗死体积均较为稳定,且100个栓子数量组的梗死面积要大于50个栓子数量组.结论 改进的微栓子栓塞法构建大鼠模型的成功率较高,可重复性较好,可以为人缺血性脑卒中的研究提供重要的参考.
Objective To explore the effect of an improved method of microemboli embolism of constructing cerebral ischemia in SD rats. Methods 30 male SD rats weighing 250~300g were randomly divided into 3 groups: sham operation group, the 50 emboli group and the 100 emboli group. Rat cerebral ischemia models was constructed with the improved microemboli embolism method. After 24h of the construction, neurological score would be evaluated, followed by the TTC staining to calculate the infarct volume. Results Neurological score of the sham operation groups was 0, while the 50 emboli group and 100 emboli group were closed to 2 points, the average score of 100 emboli group was higher than the 50 emboli group. The volume of cerebral infarction in sham operation group was 0, while the volume of cerebral infarction in 50 emboli group and 100 emboli group were relatively stable, and the infarction area of 100 emboli group was greater than 50 emboli group. Conclusion The improved microemboli embolism method has a high success rate and good repeatability, which provide important reference value for the study of ischemic stroke in human.
