槐耳醇提取物通过抑制胃癌细胞周期影响胃癌增殖的研究 被引量:1

摘要 目的 探究槐耳醇提取物通过抑制胃癌细胞周期影响胃癌增殖的作用.方法 利用CCK-8法检测不同浓度槐耳醇提取物对AGS、MGC-803胃癌细胞的增殖抑制作用;利用划痕实验验证槐耳醇提取物作用对AGS和HGC-27胃癌细胞的侵袭能力的影响;流式细胞术检测相同浓度的槐耳醇提取物和槐耳清膏对MGC-803胃癌细胞的凋亡情况;Western blot法检测相关周期蛋白;此外,对比验证槐耳醇提取物和槐耳清膏分别对MGC-803胃癌皮下瘤裸鼠模型的作用.结果 槐耳醇提取物对胃癌细胞AGS和MGC-803具有明显增殖抑制作用,并呈现剂量依赖性;槐耳醇提取物作用胃癌细胞后,其迁移能力减弱也具有时间依赖性(P<0.05),随槐耳醇提取物的作用时间的增加其迁移能力下降明显.使用0.1、0.2、0.3mg/ml的槐耳醇提取物处理MGC-803胃癌细胞,细胞中出现的凋亡细胞死亡量分别为45.3%、55.6%和66.8%;0.05mg/ml的槐耳醇提取物和槐耳清膏分别作用MGC-803细胞后,两组细胞周期蛋白CyclinD1明显低于空白对照组,且槐耳醇提取物组低于槐耳清膏组;槐耳醇提取物灌胃对荷瘤小鼠的抑制作用也显著,与空白对照组和槐耳清膏组相比,使用槐耳醇提取物的荷瘤小鼠肿瘤大小明显减小(P<0.05).结论 在细胞实验中表明槐耳醇提取物可以抑制周期蛋白CyclinD1影响胃癌细胞的增殖,在胃癌细胞实验和荷瘤小鼠模型中可以看出相同浓度的槐耳醇提取物比槐耳清膏具有更强的抗肿瘤增殖的作用. Objective To explore the effect of ethanol extract of huaier on proliferation of gastric cancer through inhibiting cell cycle of gastric cancer. Methods Human gastric cancer cell AGS,MGC-803 were treated with huaier ethanol extract of different concentrations in vitro.CCK-8 assay was used to assay the growth of gastric cancer cells. Scratch experiments were performed to evaluate the invasion ability of AGS and HGC-27 gastric cancer cells by using ethanol extract of huaier. Flow cytometry was performed to analyze apoptosis rate. Western blot was used to detect the expression of related cycle proteins. The effect of ethanol extract of huaier on MGC-803 gastric cancer subcutaneous tumor model was tested in nude mice. Results The ethanol extracts of huaier had significant inhibitory effects on the proliferation of gastric cancer cell AGS and MGC-803 in a dose dependent manner. Gastric cancer cells was treated with huaier ethanol extract,its migration ability was weakened in a time dependent manner(P<0.05). The effect time of huaier ethanol extract increased its migration ability decreased. With the increase of extract concentration,G1 phase cell arrest and apoptosis rate also increased. The MGC-803 gastric cancer cells were treated with ethanol extracts of 0.1,0.2 and 0.3mg/ml,and the apoptotic cell death in the cells were 45.3%,55.6% and 66.8%,respectively. After treatment with the same concentration of 0.05 mg/ml huaier ethanol extract and huaier water extract,the CyclinD1 in the two groups was significantly lower than that in the control group. In addition,huaier ethanol extract were lower than those huaier water extract. The inhibitory effect of extract of huaier on nude mice was also significant(P<0.05). Compared with the control group,the tumor size of nude mice using the extract of huaier was significantly reduced. Conclusion It was shown in cell experiments that ethanol extracts of huaier can inhibit the proliferation of gastric cancer cells induced by Cyclin D1. It also can be seen in gastric cancer cell experiments and tumor-bearing mouse models that at the same concentration,huaier ethanol extract has more strong anti-tumor proliferation effect than huaier water extract.
