To determine the role of the endogenous MeJA levels in the development of plants, transgenic soybean [Glycine max (L.) Merrill] plants harboring NTR1 gene encoding for jasmonic acid carboxyl methyltransferase (JMT) were produced. The activation of NTR1 gene expression resulted in the production of MeJA. Overexpression of the NTR1 cDNA under the regulation of cauliflower mosaic virus (CaMV) 35S promoter in the transgenic soybean plants was confirmed using Northern blot analysis. The significant differences in leaf and root growth patterns were observed between the transgenic plants and the wild-type plants. The leaves of the transgenic plants were slightly elongated in length but dramatically narrowed in width compared with the nontrans-formed wild-type plants. In addition, elongation of primary root was inhibited in the overexpressed transgenic soybean plantlets, whereas the development of lateral root was stimulated relative to the nontransformed plants. The leaves of the transgenic plants showed 2-2.5-fold higher levels of MeJA than the control plants. These results indicated that the increased endogenous levels of MeJA is involved in regulation of morphogenesis in soybean plants.展开更多
AIM: To investigate the implication of the hypoxia inducible factor HIF-1α mRNA in gastric carcinoma and its relation to the expression of vascular endothelial growth factor (VEGF) protein, tumor angiogenesis inva...AIM: To investigate the implication of the hypoxia inducible factor HIF-1α mRNA in gastric carcinoma and its relation to the expression of vascular endothelial growth factor (VEGF) protein, tumor angiogenesis invasion/metastasis and the patient's survival. METHODS: In situ hybridization was used to examine expression of HIF-1α mRNA, and immunohistochemical staining was used to examine expression of VEGF protein and CD34 in 118 specimens from patients with gastric carcinoma. RESULTS: The positive rates of HIF-1α mRNA and VEGF protein were 49.15% and 55.92%, respectively. Positive expressions of HIF-1α and VEGF in stage T3-T4 tumors and those with vessel invasion, lymph node metastasis and distant metastasis were dramatically stronger than stage T1-T2 cases and those without vessel invasion, lymph node metastasis and distant metastasis. The mean microvascular density (MVD) in stage T3-T4 tumors and those with vessel invasion, lymph node metastasis and distant metastasis was significantly higher than stage T1-T2 tumors and those without vessel invasion, lymph node metastasis and distant metastasis. The mean MVD in tumors with positive HIF-1α and VEGF expression was significantly higher than that in tumors with negative HIF-1α and VEGF expression. The expression of HIF- 1α was positively correlated with VEGF protein. There were positive correlations between MVD and expression of HIF-1α and VEGF. The mean survival time and the S-year survival rate in cases with positive expression HIF-1α and VEGF and MVD value ≥ 41.5/0.72 mm^2 were significantly lower than those with negative expression of HIF-1α and VEGF and MVD value 〈 41.5/0.72 mm^2. CONCLUSION: Overexpression of HIF-1α is found in gastric carcinoma. HIF-1α may induce the angiogenesis in gastric carcinoma by upregulating the transcription of VEGF gene, and take part in tumor invasion and metastasis. To reduce greenhouse gas emissions from fossil fuel fired power plants,a range of new combustion technologies are being developed or refined,including oxy-fuel combustion,co-firing biomass with coal and fluidized bed ...To reduce greenhouse gas emissions from fossil fuel fired power plants,a range of new combustion technologies are being developed or refined,including oxy-fuel combustion,co-firing biomass with coal and fluidized bed combustion.Flame characteristics under such combustion conditions are expected to be different from those in normal air fired combustion processes.Quantified flame characteristics such as temperature distribution,oscillation frequency,and ignition volume play an important part in the optimized design and operation of the environmentally friendly power generation systems.However,it is challenging to obtain such flame characteristics particularly through a three-dimensional and non-intrusive means.Various tomography methods have been proposed to visualize and characterize flames,including passive optical tomography,laser based tomography,and electrical tomography.This paper identifies the challenges in flame tomography and reviews existing techniques for the quantitative characterization of flames.Future trends in flame tomography for industrial applications are discussed.展开更多

This information is of relevance for in situ alpha spectrometry from d...The response of a silicon alpha detector to beta particles, electrons and photons was investigated using measurements and Monte Carlo simulations. This information is of relevance for in situ alpha spectrometry from different surfaces at ambient air pressure. According to the simulations, photon detection efficiencies were more than two orders of magnitude smaller than those of electrons. Photons generate signals mainly by Compton electrons. Counts originating from beta particles, electrons and photons were usually below 1 MeV in energy and no clear peaks could be identified from the measured spectra. Unequivocal identification of radionuclides emitting beta particles, electrons and photons is not possible when a mixture of different radionuclides is present in the source. However, radionuclide classification according to their emission energies appears to be possible. Surface contamination measurements will benefit from this capability.展开更多
Objective:To quantitatively detect the expression level of PRL-2 in primary hepatocellular carcinoma using real-time fluorescence quantitative PCR.Methods:Total RNA isolated from human HCC and liver tissue adjacent to...Objective:To quantitatively detect the expression level of PRL-2 in primary hepatocellular carcinoma using real-time fluorescence quantitative PCR.Methods:Total RNA isolated from human HCC and liver tissue adjacent to the tumor was reversely transcribed into cDNA.Real-time fluorescence quantitative PCR(Q-PCR) method was used to analyze the expres-sion level of PRL-2 gene.Results:The Q-PCR method was performed successfully to precisely detect RNA level.PRL-2 was expressed in all portal vein tumor thrombosis(PVTT) and HCC,but only in some paratumor tissue.The highest expression level of PRL-2 gene was recorded in PVTT;meanwhile expression level of PRL-2 was higher than that in paratumor liver tis-sues and in HCC(P < 0.01),and it was higher in HCC than that in paratumor liver tissues.Conclusion:The Q-PCR may be the most precise method to quantitatively detect RNA level and can be used in gene expression changes.The PRL-2 gene has higher expression in PVTT than that in HCC and in paratumor liver tissue cells,indicating that it plays an important role in the development and metastasis of the HCC.展开更多

With studies abroad and our preliminary investigation as base, this study developed a scale to test the factors SRL includes. According to the result of 905 teachers working in elementary school by exploratory factor analysis, the SRL for elementary teachers is consisted of its sociality (it included selecting leads and seeking for instructing), its motivation(it included self-improvement and self-excelling), its methods(it included strategy use and habitual behavior), and its outcomes(it included extensive reading and teachers' professional development). All the result indicated Teachers' SRL Scale had clear factor structure, good reliability and validity. It can be used to test the current operating situation of SRL for teachers working in elementary school.展开更多

This study therefore aims at evaluating the combined effects of cassava peel compost with poultry manure and mineral fertilizer on growth and yields of Corchorus olitorius. Pots (factorial laid out in a randomized complete block design) and field (split-plot with fertilizer being the main effect and the varieties the sub plot effect) experiments were carried out during rainy season at Ibadan, Nigeria. Treatments consisted of 0, 5, l0 and 20 t/ha composts prepared from cassava peel and poultry manure in ratio 3:1 and mineral fertilizer (NPK 15:15:15) at the rate of 250 kg/ha, using two varieties of C. olitorius. Five and six weeks after planting (WAP), plants were assessed for growth and yields in pot and field experiments respectively, and also in residual studies. Application of compost significantly improved growth (heights) (36.50-41.56 cm), while morphological and dry weight values increased with increase in compost. Marketable yield increase of 13.2%, 22.3%, 32.1% and 29.3% were recorded using 5, 10, 20 t/ha compost and NPK fertilizer respectively in the field. Corresponding increase when ratooned were 40.0%, 42.9%, 57.1% and 52.0%, respectively. Application of compost at the rate of 20 t/ha performed better in the main and residual effect experiments compared to NPK fertilizer and other compost rates. Application of cassavas peels compost at the rate of 20 t/ha proved to be adequate for optimum growth and yield performance of C. olitorius.展开更多

With the increasing concentration of As (III), the xylanase activity elevated first then dropped in seed, but it descended first then ascended in root and plumule. As the sampling time prolonged, the xylanase activity of seeds climbed first then dropped on the four as (III) concentration, the same trend also appeared in pulume, as the as (Ill) concentration went up, the xylanase activity moved up simultaneity.
