The whole liver of a mouse was removed from the body, homogenated, and extracted by ethyl acetate. The organic layer was isolated and evaporated to dryness, the residue was reconstituted in 0.2 mL mobile phase for centrifugation, and 50 uL of the supernatant was injected into the/-IPLC instrument. The sample was separated on a Shimadzu ODS column (150 mm × 4.6 mm, 5 um) at 35 ℃ and detected by ultraviolet (UV) detector at the wavelength of 305 nm. The mobile phase consisted of methanol and 0.1 mol/L acetic acid (4:6, v/v) with the flow-rote at 1 mL/min. The limit of detection was 3.0 ng/g in liver homogenate with a signal/noise ratio of 3:1. The linear range of the calibration curve was 5.0-120.0 ng/g. The mean recoveries at the concentrations of 6, 10 and 80 ng/g were 102%, 96.0% and 91.5%, respectively. The RSDs for inter- and intra-day assays were less than 5%. Compared with other reported methods, this method was faster and more sensitive. It was also proved to be of good linearity, selectivity, accuracy and precision, and can be efficiently applied to the pharmacoldnetic study of trans-resveratrol in mouse liver.展开更多

Actin microfilaments could be visualized only in the late_developmental stage of the immature pollen. During this developmental stage, microfilaments, initially composed of very short fibrils, develop into a very complex and novel network that sometimes totally and sometimes partially encloses the vegetative nucleus and the spherical shaped generative cell in the central cytoplasm of the immature pollen. The behavior of the actin microfilamentous structure throughout the late_developmental stage of the immature pollen is extremely dynamic, and the likelihood of this structure in generating forces for vegetative nucleus and generative cell movement in the immature pollen has been discussed. No actin filaments were visualized in the spherical generative cells.展开更多

METHODS Transplanted tumor tissue was used for in vitro primary culture of U14 cervical carcinoma cells. After 20 passages, we examined its morphology, chromosomes, tumorigenicity and produced a growth curve. CK was detected by immunohistochemistry, the cell cycle determined by flow cytometry and the metastatic potential assessed in 615 and C57BL/c mice. We also transfected the cells with the pEGFP-N1 plasmid. RESULTS A newly established murine cell line was passaged 50 times over a period of 10 months. The cells grow as a partially suspended culture, and are immunohistochemically CK(+). The cell line is characterized by a hypotetraploid karyotype, a chromosomal number of 64-68 and a doubling time of 21.8 h. Exponential growth occurs by the third and forth day of culture. Cell cycle analysis showed G1 34%, G2 26%, and 40% in the S phase. The tumorigenicity was 100% upon implantation. No mycoplasma contamination was detected. A monoclonal continuous U14-GFP cell strain which was 100% GFP (+) was also produced. CONCLUSION We successfully established a new murine cervical U14 carcinoma cell line and an U14-GFP monoclonal strain. These cell lines are ideal for combined in vivo and in vitro tumor research.展开更多

When the methane volume fraction is 10M, the maximum explosion pressure is 0. 264 MPa and the injury is the most serious. Specially, some designed obstacles put in the open space pipeline are conducive to producing more stronger gas explosion shock wave. Accordingly, the injury effect of methane explosion on mouse is enhanced under obstacles condition. When the methane volume fraction is 10%, the maximum explosion pressure can reach 0. 298 MPa under obstacles conditiorL It can be concluded that to reduce explosive accident impact, the obstacles in coal mine should be avoided. With the explosions increasing, the death pressure of mouse decreases.展开更多

The levels of 6-and 12-month-old C57 mice were much lower compared with that of the l-month-old mouse. However, there was no significant difference between these two groups. This result suggests that gene transcription level of hippocampus β NGF in a C57, mouse descends evidently as it grows up. Base T at Site 294 of β NGF cDNA was substituted by C in the C57, mouse, as revealed by DNA sequencing for the first time. Nevertheless, this polymorphism of nucleotide did not make any difference in amino acid composition.展开更多

Although most oocytes went through GVBD 2-4 hrs after culture, 13.6% remained at GV stage 8 hrs after culture.TEM observation revealed that nucleoli of oocytes which failed to go through GVBD were composed of fibrillar-granular component, small vacuoles and fibrillar centers or showed small vacuoles on nuclear surface. During GVBD, the nucleoli became smaller and smaller and finally disappeared with the nuclear-associated chromatin dislocated to the periphery. Nuclear membrane with attached chromatin became folded and electronic dense cores appeared in the center of chromatin clumps at the same time. The last event of GVBD was the disruption of nuclear membrane. At the end of the 5th hr after culture, meiosis progressed to prometaphase I.Chromosomes, distributed in the original GV area free of organelles, were surrounded by large quantity of mitochondria and small SER vesicles. At the end of the 12th hr after culture,48. 1% of the oocytes emitted PB1.Decondensing sperm head and early male pronuclcus(mPN)with condensed nucleoli were found 1-2 hrs after insemination. The formation and enlargement of female PN(fPN) occurred a little earlier than that of mPN. 33.3% finished syngamy at 8-9 hrs after insemination. The process of nucleolus formation was reverse to that in GVBD. The oolemma modification caused by cortical reaction could effectively inhibit polyspermy.in contrast, there were sperm binding to the oolemma where CGs failed to be released. In addition, PB2 was emitted 2-5 hrs after insemination. The difference between PB1 and PB2 as well as the abstriction of polar body were also discussed.展开更多

文摘BACKGROUND ATP sensitive K+(K_(ATP))channels are ubiquitously distributed in various of cells and tissues, including the liver. They play a role in the pathogenesis of myocardial and liver ischemia.AIM To evaluate the radiation-induced changes in the expression of K_(ATP)channel subunits in the mouse liver to understand the potential role of K_(ATP)channels in radiation injury.METHODS Adult C57BL/6 mice were randomly exposed toγ-rays at 0 Gy(control,n=2),0.2 Gy(n=6),1 Gy(n=6),or 5 Gy(n=6).The livers were removed 3 and 24 h after radiation exposure.Hematoxylin and eosin staining was used for morphological observation;immunohistochemical staining was applied to determine the expression of K_(ATP)channel subunits in the liver tissue.RESULTS Compared with the control group, the livers exposed to 0.2 Gyγ-ray showed an initial increase in the expression of Kir6.1 at 3 h,followed by recovery at 24 h after exposure.Exposure to a high dose of 5.0 Gy resulted in decreased expression of Kir6.1 and increased expression of SUR2B at 24 h.However, the expression of Kir6.2,SUR1,or SUR2A had no remarkable changes at 3 and 24 h after exposure to any of these doses.CONCLUSION The expression levels of Kir6.1 and SUR2B in mouse liver changed differently in response to different radiation doses, suggesting a potential role for them in radiation-induced liver injury.
文摘In order to study the effect of weak noise on the sound signal extraction of mouse (Mus musculus Km) inferior collicular (IC) neurons from environments,we examined the changes in frequency tuning curves (FTCs) of 32 neurons induced by a weak noise relative to 5 dB below minimum threshold of tone (reMT-5 dB) under free field stimulation conditions. The results were as follows:① There were three types of variations in FTCs,sharpened (34.4%),broadened (18.8%),and unaffected (46.9%),nevertheless,only the alteration of sharpened FTCs was statistically different.② Sharpness of frequency tuning induced by a reMT-5 dB noise was very strong.Q 10 and Q 30 of FTCs were increased by (34.42±17.04)% (P=0.026,n=11) and (46.34±22.88)% (P=0.009,n=7).③ The changes of inverse-slopes (ISs,kHz/dB) between high (IS high) and low (IS low) limbs of FTCs were dissymmetry. The IS high of FTCs decreased markedly (P=0.046,n=7),however,there was little change (P=0.947,n=7) in IS low.Our data revealed for the first time that the weak noise could sharpen frequency tuning and increase the sensitivity on the high frequency of sound signal in IC neurons of mouse.

文摘To develop a sensitive high performance liquid chromatography (HPLC) assay for the determination of trans-resveratrol in mouse liver. It was also proved to be of good linearity, selectivity, accuracy and precision, and can be efficiently applied to the pharmacoldnetic study of trans-resveratrol in mouse liver.
文摘Green fluorescent protein (GFP) fused to the F_actin binding domain of mouse talin labels the actin cytoskeleton in the immature pollen of stable transformed rice (Oryza sativa L.) plants. No actin filaments were visualized in the spherical generative cells.
文摘[ Objective] The aim of this study is to construct the model for simple obesity induced by high-fat diet, which is closest to human obesity, laying a foundation for the studies of obesity related theories. [Method[ ICR and KM mice, half male and half female, were randomly divided into the high-fat diet experimental group and the normal diet control group based on body weights, and certain days later, body weight, Lee' s index, wet weight of adipose tissue, quantity of adipose cell in the same visual field and blood indices were measured. [Result]All indices mentioned a- bove of the female I CR mouse had significant statistical differences with those of the control group (P 〈 0.01 or P 〈 0.05). [ Conclusion] To con- struct mouse nutritional obesity model successfully, different high-fat diets are required by different lines as well as different sexes in the same line.
文摘OBJECTIVE To establish a murine uterine cervical cancer cell line and to define its biological characters. These cell lines are ideal for combined in vivo and in vitro tumor research.
文摘The killing and injury effects of gas explosion shock wave on mouse in an open space pipeline is tested experimentally. With the explosions increasing, the death pressure of mouse decreases.
文摘β-Nerve growth factor (β NGF) mRNA levels in hippocampa from C57BL/6J mice of different ages were compared by semi-quantitative RT-PCR method, taking β-actin gene as an internal control. Nevertheless, this polymorphism of nucleotide did not make any difference in amino acid composition.
文摘The chronological and morphological changes of the nucleus during mouse oocyte maturation and fertilization were systematically studied. The difference between PB1 and PB2 as well as the abstriction of polar body were also discussed.
文摘The effect of gonadal hormones on brain tubulin synthesis has been studied in the castrated middle-aged male mouse and testes intact oldaged male mouse.Estrogen exerts potent influences on stimulating tubulin synthesis.Treatment with estradiol benzoate or estradiol benzoate and hydroxyprogesterone caproate increase the amount of tubulin by almost 37%-43%.Administration of testosterone propionate can produce a 13%-23% increase. Both estrogen and androgen can stimulate tubulin synthesis after the critical period of brain development but, estrogen plays a stronger role than androgen in stimulating tubulin synthesis in male mouse.
